Project description:Female mosquitoes require a blood meal for oogenesis, and thereby receive a substantial iron load in the forms of holo-transferrin and hemoglobin. Our previous data showed that during digestion of a blood meal, the gut iron concentration decreases 10-fold, while that of the ovaries doubles from ingestion to 72 hours post feeding. Approximately 72 hours post feeding, eggs are laid with ~125 ng Fe each. We are interested in the effects of the blood meal on the expression of iron related proteins detected in the ovaries during time post feeding before eggs are laid. We have used shotgun proteomic analysis to identify proteins in the developing ovaries of Aedes aegypti; this information provides further insight into the effect of a blood meal on mosquito oogenesis.

Project description:Rockefeller and Singapore strain Aedes aegypti female mosquitoes differ in the number of bacteria present in the midgut. Females from each strain were either maintained on 3% sucrose solution, fed a sterile blood meal, or fed a blood meal containing a cocktail of bacteria. Differential transcript abundance was compared between females from each strain/treatment combination and a common reference sample pool. The overall goal was to determine how gene expression in Rockefeller females differs from Singapore females in order to better understand why the gut microbiome differs between the strains.

Project description:Female Aedes aegypti mosquitoes impose a severe global public health burden as primary vectors of multiple viral and parasitic pathogens. Under optimal environmental conditions, Aedes aegypti females have access to human hosts that provide blood proteins for egg development, conspecific males that provide sperm for fertilization, and freshwater that serves as an egg-laying substrate suitable for offspring survival. As global temperatures rise, Aedes aegypti females are faced with climate challenges, like intense droughts and intermittent precipitation, which create unpredictable and suboptimal conditions for the egg-laying step of their reproductive cycle. Aedes aegypti mosquitoes nonetheless show remarkable reproductive resilience, but how they achieve this is unknown. Here we show that under drought-like conditions simulated in the laboratory, mated, blood-fed Aedes aegypti females carrying mature eggs retain them in their ovaries for extended periods, while maintaining the viability of these eggs until they can be deposited in freshwater. Using transcriptomic and proteomic profiling of Aedes aegypti ovaries, we identify two previously uncharacterized genes – here named tweedledee and tweedledum – that show ovary-enriched, temporally-restricted expression during egg retention. These genes are mosquito-specific, linked within a syntenic locus, and rapidly evolving under positive selection, raising the possibility that they serve an adaptive function. Using loss-of-function mutagenesis to disrupt both genes, we show that, tweedledee and tweedledum, which encode secreted proteins, are specifically required for extended retention of viable eggs, such as during intermittent precipitation or drought. These results highlight an elegant example of taxon-restricted genes at the heart of an important adaptation that equips Aedes aegypti females with “insurance” to, when contextually appropriate, flexibly extend their reproductive sequence without losing reproductive capacity, thus allowing this species to exploit diverse and unpredictable/chaotic/changing habitats.

Project description:Aedes aegypti mosquito, vector for several viral diseases, undergoes significant physiological changes after a blood meal. Through single-cell RNA sequencing and metabolomics, we unveiled dynamic cellular composition and metabolic adaptations within abdominal midgut and fat body tissues. We revealed high cell diversity, specialized in digestion, metabolism, immunity, and reproduction. While the midgut primarily comprises enterocytes, enteroendocrine, cardia and intestinal stem cells, the fat body consists of not only trophocytes and oenocytes, but also a substantial population of hemocytes and fat body-yolk cells (FYC). The fat body, exhibiting a more complex metabolomic profile than the midgut, played a central role in immune and metabolic gene expression, particularly within trophocytes and FYCs. Additionally, insect-specific viruses were detected at the single-cell level, mainly in the midgut at later stages post-blood meal. These findings offer new vector control strategies by targeting specific abdominal cell populations and metabolic pathways involved after a blood meal.

Project description:Aedes aegypti [Linnaeus in Hasselquist (Diptera: Culicidae); yellow fever mosquito] transmits several viruses that infect millions of people each year including, Zika, dengue, yellow fever, chikungunya, and West Nile. Disease transmission occurs during blood feeding. Only the females blood feed as they require a bloodmeal for oogenesis. In the bloodmeal, females receive a substantial iron load in the forms of holo-transferrin and hemoglobin. We are interested in the effects of the iron in a bloodmeal on the expression of proteins during oogenesis. Our previous data showed that during digestion of a blood meal, the gut iron concentration decreases 10-fold, while that of the ovaries doubles from ingestion to 72 hours post feeding. Approximately 72 hours post feeding, eggs are laid with ~125 ng Fe each. We are interested in the effects of the blood meal iron on the expression of proteins detected in the ovaries during the early oogenesis, 24 hours post feeding, before eggs are laid. We have used tandem mass tag-labeling proteomics to quantify proteins expressed at this early stage following feeding of a controlled iron diet. Our findings provide the first quantitative report of differential ovary protein expression in early oogenesis in mosquitoes fed three different iron diets.

Project description:Oral susceptibility of Aedes aegypti mosquitoes to dengue viruses varies between different Aedes species and strains. However, the midgut-specific transcriptional profile that may produce this variation is presently obscure and was the subject of our investigation. The variation in active expression between dengue-2 susceptible (SUS) and refractory (REF) mosquitoes was investigated during the first critical 96 hours after infection Transcriptional profiles were mined from respective guts using the serial analysis of gene expression technique (SAGE) and libraries constructed from midguts obtained from mosquitoes that received a dengue-2 infected blood meal (DENV-2), a non infected blood meal (naive) or a 5% sucrose meal (SM). Here we report that variation between DENV-2 infected libraries versus respective naïve libraries revealed very few transcripts that were common and statistically significant in DENV-2 infected libraries. In addition, the expression profiles among libraries displayed up regulation of antisense transcripts especially in the SUS strain. A strong proclivity towards strain-specificity in differential expression was observed, which suggested an exclusive transcription that is likely up-regulated after DENV-2 infection Thirty Aedes aegypti female mosquitoes aged 4-5 days were transferred to 500 ml paper cups and offered a 5% sucrose meal (SM), a naïve blood meal or a dengue-2 (JAM 1409 strain) infectious blood meal, using standard artificial membrane feeders. Project description:We used our deep sequence data and bioinformatics to analyzed the miRNA repertoires expressed in the CA of pupae, sugar-fed and blood-fed female Aedes aegypti mosquitoes. In total, 156 mature miRNAs were detected in the CA, with 84 displaying significant differences in expression among the three CA developmental stages. Notably, the changes in the miRNA repertoire in the CA in the pupa-adult transition have completely different characteristics compared with the changes from sugar-fed to blood-fed mosquitoes.
