Hormone effects on gene expression in somatic cells of rat testes (Rattus norvegicus)

ABSTRACT: Analysis of hormone effects on irradiated LBNF1 rat testes, which contain only somatic cells except for a few type A spermatgogonia. Rats were treated for 2 weeks with either sham treatment (group X), hormonal ablation (GnRH antagonist and the androgen receptor antagonist flutamide, group XAF), testosterone supplementation (GnRH antagonist and testosterone, group XAT), and FSH supplementation ((GnRH antagonist, androgen receptor antagonist, and FSH, group XAFF). Results provide insight into identifying genes in the somatic testis cells regulated by testosterone, LH, or FSH.

Identification of Rat foreskin biomarkers of in utero dibutyl phthalate exposure

Project description:Dibutyl phthalate was administered to pregnant Sprague Dawley rats from gestational days 16-20 at either a 100 mg/kg/day or 500 mg/kg/day dose level. This timeframe covers the reproductive masculinization window which corresponds to increased androgen signalling. Dibutyl phthalate has been shown to disrupt testosterone production leading to male reproductive abnormalities. As such, we selected this exposure window for our study and examined gene expression changes in the male rat foreskin, which expresses the androgen receptor. We collected tissue samples at both gestational day 20 to identify gene expression changes immediately after exposure, and postnatal day 5 to identify gene expression changes persisting after birth using microarray analysis (Illumina RatRef 12 Bead Chips). To determine whether gene expression changes were brought on by decreased androgen signalling or additional effects of dibutyl phthalate exposure, we exposed rats to the potent androgen receptor antagonist flutamide (5 mg/kg/day) during the same period of development. Gene expression changes were compared to determine which were brought on by disruption of androgen signalling and which were the result of other aspects of chemical exposure. The flutamide exposure study consisted of seven control dams administered corn oil and seven dams treated with 5 mg/kg/day flutamide. Two foreskin samples per litter were pooled for gene expression microarray analysis using the Affymetrix Gene 1.0 ST Array.

Gene expression analysis of bovine oocytes at optimal coasting time combined with GnRH antagonist during the no-FSH period

Project description:The objective of the study was to analyze the effect of a GnRH antagonist used during the FSH withdrawal period (coasting) on transcriptome changes in in vivo bovine oocytes at the GV stage. Oocytes were collected from super-stimulated animals after a coasting duration of 68hrs (optimal condition, control) and also from the same animal who received the GnRH antagonist (treated) during the whole coasting period. 68hrs of coasting (control) was compared with the 68hrs + GnRH antagonist (treatment), for each cow individually. Overall, 6 hybridizations were done, corresponding to the three biological replicates and one comparison using a dye-swap set-up.

Identification of androgen-responsive genes in fish testis

Project description:Sexual steroids play an important role in gonad maturation in all vertebrates. However, the molecular mechanisms regulating of spermatogenesis in seasonal breeding species are yet poorly understood. In this study we investigated the effects of testosterone and 11-ketotestosterone (11KT) - a teleost fish specific androgen - on testicular gene expression at the onset of puberty in the rainbow trout, Oncorhynchus mykiss. Rainbow trout 13 months old were used. Immature males (spermatogonia only, stage I-II) were submitted to androgen supplementation by hormone implants for 7 or 14 days. This included : - untreated animal controls (N=14) - animals subjected to testosterone supplementation (0.1mg) for 7 days (N=5) - animals subjected to testosterone supplementation (0.2mg) for 7 days (N=4) - animals subjected to testosterone supplementation (0.2mg) for 14 days (N=5) - animals subjected to 11-ketotestosterone supplementation (0.25mg) for 7 days (N=7). Individual testes were recovered for microarray experiments and histological analyses. Circulating levels of corresponding androgens were also determined.

COX-2 network as predictive molecular marker for clinical pregnancy in IVF

Project description:CONTEXT Nowadays, the molecular mechanisms involved in endometrial receptivity and implantation are still not clear. OBJECTIVE The gene expression of human endometrium of patients undergoing an IVF treatment with GnRH antagonists/rec-FSH was studied. CONCLUSIONS COX-2 has been extensively studied as a crucial fertility element in both knock-out mice and human. It appears that increased expression of COX-2 and/or SCGB1D2 on the day of oocyte retrieval in GnRH antagonist/rec-FSH stimulated cycles coincides with a lower probability of achieving a clinical pregnancy in this cycle. Premature progesterone rise

Project description:Premature progesterone (P) rise during GnRH antagonist cycles for IVF is a frequent phenomenon and has been associated with lower pregnancy and implantation rates. Different thresholds of progesterone have been used so far to define its premature rise during the follicular phase of an IVF stimulated cycle. In this study, we evaluated endometrial gene expression on the day of oocyte retrieval according to the level of serum progesterone on the day of hCG administration in GnRH antagonist cycles.Endometrial biopsies from eleven patients were taken with a Pipelle de Cornier (Prodimed, Neuilly-en-Thelle, France) on the day of oocyte retrieval in a GnRH antagonist/rec-FSH stimulated IVF cycle with fresh embryo transfer. Biopsies were analysed for gene expression with Affymetrix Human Genome (HG) U133 Plus 2.0 Arrays and GCOS software (Affymetrix, Santa Clara, CA, USA). Patients were divided into three different groups according to their progesterone serum concentration on the day of hCG administration (A) P <= 0.9 ng/mL, (B) 1 < P < 1.5 ng/mL, and (C) P > 1.5 ng/mL. Serum P was measured with the automated Elecsys immunoanalyser (Roche Diagnostics, Mannheim, Germany). Selected differentially expressed genes were validated with quantitative real-time PCR (QPCR) with TaqMan Gene Expression Assays (Applied Biosystems, Foster City, CA, USA).
