Plasma Level of Circular RNA hsa_circ_0000190 Correlates with Tumor Progression and Poor Treatment Response in Advanced Lung Cancers


ABSTRACT: Circular RNAs (circRNAs) were reported to play roles in cancers by targeting pro- and anti-oncogenic miRNAs. However, the mechanisms of circRNAs in lung cancer progression and their prognostic value of treatment response remain unclear. This study used next generation sequencing (NGS) to analyze the transcriptomes and circRNA profiles of 2 lung cancer cell lines (A549 and HCC827) and 1 normal lung cell line (BEAS-2B), and identified the highly overexpressed hsa_circ_0000190 and hsa_circ_000164 as potential biomarkers. By using highly sensitive RT-ddPCR method, these circRNAs were shown to be secreted by cell lines and were detected in human blood. We predicted the potential miRNA targets of the two circRNAs, and the further mRNA targets of these miRNAs. This NGS data allows us to compare the levels of predicted mRNAs to delineate potential involving mechanisms of the two circRNAs.
