Expression data from DE-71 treated rats at PND4

ABSTRACT: To identify liver transcripts differentially expressed between control samples and animals exposed to DE-71, we collected RNA from male pups at postnatal day 4 (PND4) after the Wistar Han dams were exposed to 0, 0.1, 15, or 50 mg/kg DE-71. These samples were interrogated with the Affymetrix Rat Genome 230 2.0 GeneChip array A total of 0, 15, and 162 gene transcripts were differentially expressed between control samples and 0.1, 15, and 50 mg/kg DE-71, respectively (using a false discovery rate (FDR) threshold of 0.05). These transcripts were mapped to 0, 13, and 161 genes using Ingenuity Pathway Analysis.

Project description:To identify liver transcripts differentially expressed between control samples and animals exposed to PBDE-47, we collected RNA from male pups at postnatal day 4 (PND4) after the Wistar Han dams were exposed to 0, 0.1, 15, or 50 mg/kg PBDE-47. These samples were interrogated with the Affymetrix Rat Genome 230 2.0 GeneChip array. A total of 7, 50, and 149 gene transcripts were differentially expressed between control samples and 0.1, 15, and 50 mg/kg PBDE-47 (using a false discovery rate (FDR) threshold of 0.05). These transcripts were mapped to 2, 40, and 139 genes using Ingenuity Pathway Analysis.

Rapid genomic changes induced by PTH, SIK-inhibition, FGF23, and 1,25(OH)2D3 in the mouse kidney

Project description:In vivo ChIP-seq in mouse kidney: we examine the genomic recruitment of VDR, RXR, pCREB, CBP, CRTC2 (TORC2), SRC1, SRC3, BRD4, RNApolII, H3K9ac, H3K27ac, and H3K36me3 after intraperitoneal injection of 10 mg/kg body weight (bw) 1,25(OH)2D3 (in propylene glycol), 230 mg/kg bw PTH (1–84) (in phosphate buffered saline (PBS)), 50 mg/kg bw FGF23 (in PBS + 0.1% BSA), 30 mg/kg YKL-05-099 (PBS + 25mM HCl), 40 mg/kg SK-124 (15% HPBCD (hydroxypropyl β-cyclodextrin)), or vehicle (EtOH, PBS, or HPBCD). YKL-05-099 and SK-124 are salt-inducible kinase inhibitors that liberate CRTC2 from sequestration in the cytoplasm and allow CRTC2 translocation to the nucleus. Our findings show that these TFs and coactivators contribute to transcription of genes in the kidney involved in vitamin D metabolism, demonstrates SIK-inhibition as a key modulator of vitamin D metabolism, and provides molecular insight into the coordinated mechanistic actions of PTH, FGF23, and 1,25(OH)2D3 in the mouse kidney that regulate mineral homeostasis.

Project description:Global mRNA expression profiling of patient derived pancreatic carcinoma xenograft Bo63 were collected using Agilent human whole genome array (G4845A AMADID 026652, cRNA 4x44k V2) . Two different sources of RNA were analyzed: 1.) Bo63 xenograft tumors grown on nude mice treated with vehicle only as control. 2.) Bo63 xenograft tumor grown on nude mice treated with JQ1 and SAHA (SAHA 25 mg/kg 1-0-0 and JQ1 50 mg/kg 0-0-1, treatment was imitated when tumor size reached 200 mm³ +/- 20 mm³.
