Single cell gene expression profiling of the response of the dorsal motor nucleus of the vagus neurons to remote ischemic preconditioning [DMV_RIPC_RT-PCR]

ABSTRACT: We developed a mesoscale single cell transcriptomics data from hundreds of the dorsal motor nucleus of the vagus (DMV) neurons under homeostasis and following physiological perturbations. Our results revealed that homeostatic DMV neuronal states can be organized into distinguishable input-output signal processing units. Remote ischemic preconditioning induced a distinctive shift in the neuronal states towards diminishing the role of inhibitory inputs.

Project description:We developed microRNA expression profiling data from the dorsal motor nucleus of the vagus (DMV) neurons under homeostasis and following physiological perturbations of remote ischemic preconditioning and cardiac ischemia. Male Sprague-Dawley rats were subjected to remote ischemia preconditioning (rIPC), ligation of the left anterior descending artery (LAD), or rIPC with LAD following two hours later (rIPC 2h/LAD). All samples were collected 24 hours after LAD.

Neuromodulatory co-expression in cardiac vagal motor neurons of the Dorsal Motor Nucleus of the Vagus [RNAseq]

Project description:The activity of the cardiac vagal innervation is well known to be crucial to the maintenance of cardiac health, and to protect and recover the heart from injury. Only recently has this role been shown to depend on the activity of the underappreciated Dorsal Motor Nucleus of the Vagus (DMV). By combining neural tracing, transcriptomics, and 3D mapping in male and female Sprague Dawley rats we characterize cardiac-specific neuronal phenotypes in DMV. We find that the DMV cardiac-projecting neurons differentially express PACAP, CART, and synucleins, as well as evidence that they participate in neuromodulatory co-expression involving catecholamines. The significance of these findings is enhanced by previous knowledge of the role of PACAP at the heart and of the other neuromodulators in peripheral vagal targets.

Highly Selective Brain-to-Gut Communication via Genetically-Defined Vagus Neurons

Project description:The vagus nerve innervates many organs, and most if not all of its motor fibers are cholinergic. However, no one knows its organizing principles; whether or not there are dedicated neurons with restricted targets that act as "labeled lines" to perform certain functions – including two opposing ones (gastric contraction versus relaxation). By performing unbiased transcriptional profiling of DMV cholinergic neurons, we discovered seven molecularly-distinct subtypes of motor neurons. Then, by using subtype-specific Cre driver mice, we go on to show that two of these subtypes exclusively innervate the glandular domain of the stomach where, remarkably, they contact different enteric neurons releasing functionally opposing neurotransmitters (acetylcholine versus nitric oxide). Thus, the vagus motor nerve communicates via genetically-defined labeled lines to control functionally unique enteric neurons within discrete subregions of the gastrointestinal tract. This discovery provides a conceptual breakthrough on the division of labor used by the vagus motor nerve to control function.

Modulation of brain dead induced inflammation by vagus nerve stimulation

Project description:Because the vagus nerve is implicated in control of inflammation, we investigated if brain death causes impairment of the parasympathetic nervous system, hence contributing to inflammation. Brain death (BD) was induced in rats. Anaesthetised ventilated rats (NBD) served as control. Heart rate variability (HRV) was assessed by ECG. The vagus nerve was electrically stimulated (BD+STIM) during BD. Intestine, kidney, heart and liver were harvested after 6h. Affymetrix chip- analysis was performed on intestinal RNA. Quantitative PCR was performed on all organs. Serum was collected to assess TNFÎ± concentrations. Renal transplantations were performed to address the influence of vagus nerve stimulation on graft outcome. HRV was significantly lower in BD animals. Vagus nerve stimulation inhibited the increase in serum TNFÎ± concentrations and resulted in down-regulation of a multiplicity of pro-inflammatory genes in intestinal tissue. In renal tissue vagal stimulation significantly decreased the expression of E-selectin, IL1Î² and ITGA6. Renal function was significantly better in recipients that received a graft from a BD+STIM donor. Our study demonstrates impairment of the parasympathetic nervous system during BD and inhibition of serum TNFÎ± through vagal stimulation. Vagus nerve stimulation variably affected gene expression in donor organs and improved renal function in recipients. Remote ischemic preconditioning in on-pump coronary artery bypass graft surgery

Project description:In this placebo-controlled randomized controlled trial, we tested whether remote ischemic preconditioning (RIPC) elicited by four 5-minute cycles of 300 mmHg of cuff inflation/deflation of the lower limb would reduce myocardial necrosis in isoflurane-anesthetized patients undergoing on-pump coronary artery bypass graft surgery. Secondary outcomes were the perioperative release of the biomarkers NTproBNP, hsCRP, S100, atrial transcriptional profiles, and short- and long-term clinical outcomes. RIPC with concomitantly applied isoflurane did not affect the release of biomarkers or clinical outcome. NTproBNP release correlated with isoflurane- but not RIPC-induced transcriptional changes.
