Project description:The mammalian heart undergoes maturation during postnatal life to meet the increased functional requirements of the adult. However, the key drivers of this process remain poorly defined. We developed as 96-well screening platform, using human pluripotent stem cell derived cardiac organoids, to determine the molecular requirements for in vitro cardiomyocyte maturation. Here, we describe gene expression changes resulting from culturing human cardiac organoids in standard cell culture conditions and under optimized maturation conditions. We assessed our maturation conditions by comparing transcriptional changes of human cardiac organoids to RNA isolated from human heart. Interesting, analysis of these data revealed that a switch to fatty acid oxidative metabolism is a key governor of cardiomyocyte maturation and mature cardiac organoids were refractory to mitogenic stimuli.

Project description:The freshwater fish crucian carp (Carassius carassius) can survive complete oxygen depletion (anoxia) for several months at low temperatures, achieved by a combination of reduced energy demand and increased glycolysis fueled by large hepatic glycogen stores. In crucian carp, the energy-requiring protein synthesis is controlled in a tissue-specific manner when oxygen levels decrease. During anoxia, translational rates are maintained at almost normoxic levels in brain, while heart and liver translation rates are strongly reduced. However, little is known about how the global proteome of these tissues are affected by oxygen variations. By applying mass spectrometry-based proteomics, 3304 proteins in brain, 3004 proteins in heart and 2516 proteins in liver were detected, of which 66 brain proteins, 243 cardiac proteins and 162 hepatic proteins were differentially expressed during the course of anoxia-reoxygenation compared to normoxic control. The brain proteome showed few differences in response to oxygen variations, indicating that anoxic survival is not regulated through protein expression in this tissue. Cardiac and hepatic adaptions to anoxia included enrichment of mitochondrial proteins involved in aerobic respiration and mitochondrial membrane integrity. We show that enzymes in the electron transport system (ETS) are regulated in a tissue-specific manner since no ETS components were regulated in brain, but were downregulated in heart and upregulated in liver during anoxia and reoxygenation. Furthermore, complement system activation was enriched in heart during anoxia. During reoxygenation, proteins involved in the cristae junction organization were regulated in the heart, possibly explaining how reactive oxygen species can be avoided when oxygen returns in this master of anoxic survival.

The peptides were designed to be random, or pseudo-random, but fulfill requirements of immunosignaturing. This file contains data from actual 330,000 peptide arrays that used the first iteration of the peptide generation algorithm. Monoclonal antibodies were bound to the microarrays and the total number of peptides that distinguished each monoclonal was measured. This provides a baseline against which to compare purely random sequences.

Project description:In an endotoxaemic mouse model of sepsis, a tissue-based proteomics approach for biomarker discovery identified long pentraxin 3 (PTX3) as the lead candidate for inflamed myocardium. PTX3 accumulated as an octamer due to disulphide-bond formation in heart, kidney and lung ?common organ dysfunctions seen in patients with sepsis. Oligomeric moieties of PTX3 were also detectable in the circulation. The redox-state of PTX3 was quantified over the first 11 days in critically ill adult patients with sepsis. On admission day, there was no difference in the redox-state of PTX3 between survivors and non-survivors. From day 2 onwards, the conversion of octameric to monomeric PTX3 was consistently associated with a greater survival after 28 days of follow-up. For example, by day 2 post admission, octameric PTX3 was undetectable in survivors, but still constituted more than half of total PTX3 in non-survivors (P<0.001). Monomeric PTX3 was inversely associated with cardiac damage markers NT-proBNP, high sensitivity troponin I and T. In comparison to the conventional measurements of total PTX3 or NT-proBNP, the redox-sensitive oligomerization of PTX3 was more dynamic and a superior predictor of disease outcome.

Project description:Heart failure is associated with degradation of cell functions and extracellular matrix proteins, but the trigger mechanisms are uncertain. Our recent evidence in acute multiorgan failure suggests that active digestive enzymes leak out of the small intestine into the systemic circulation and cause cell dysfunctions and organ failure. Accordingly, we investigated in morning fasting plasma of heart failure (HF) patients the presence of pancreatic trypsin, a major enzyme responsible for digestion. Western analysis shows that trypsin in plasma is significantly elevated in HF compared to matched controls and their concentrations correlate with inflammatory biomarkers. The plasma trypsin levels in HF are accompanied by elevated pancreatic lipase concentrations. The trypsin has a significantly elevated activity as determined by substrate cleavage. Mass spectrometry shows that the number of proteins in the HF group is similar to controls while the number of peptides was increased about 20% in the HF patients. The peptides are derived from extracellular and intracellular protein sources and exhibit cleavage sites by trypsin as well as other degrading proteases. These results provide first evidence that active digestive enzymes leak into the systemic circulation and may participate in myocardial cell dysfunctions and tissue destruction in HF.

Project description:Thyroid hormones (THs) influence adipose tissue development and metabolism. They regulate both adipocyte proliferation and differentiation and, as they increase the metabolic rate, may be helpful for obesity treatment. However, due to their cardiotoxic effects, like tachycardia and arrhythmia, their use is limited to hypothyroid obese patients. Interestingly, some TH metabolites have been recently shown to possess the same beneficial metabolic effects as THs without any negative effect. The biogenic amine 3-Iodothyronamine (T1AM), for example, affects carbohydrate and lipid metabolism without undesirable side effects. In mice T1AM exhibits cardiac effects opposite to those associated with thyroid hormones, like bradycardia, and, in isolated working rat hearts, produces a rapid, reversible, dose-dependent decrease in cardiac output, aortic pressure and coronary flow. Intraperitoneal injections of T1AM, moreover, induce reduction of RQ from 0.9 to 0.7, both in mice and Djungarian hamsters, indicating that carbohydrate utilisation is reduced and energy requirements are covered by lipid consumption. Ketone bodies in the urine and the significant loss of body fat mass confirm that lipids are predominantly used to cover the energy requirements in response to T1AM administration. The molecular mechanisms by which T1AM favors the lipid rather than glucose catabolism are not known, but it is possible to envisage changes in gene expression, given the delayed and long-lasting phenotypic effects. The complete RQ shift, for example, is reached 4.5 h after the T1AM injection and persists at least for 24 h. In this paper we analyzed the gene expression profiles in subcutaneous adipose tissue and in liver of eight rats chronically treated with T1AM as compared with eight untreated rats. Many genes linked to lipid metabolism, adipogenesis and angiogenesis appeared modulated by T1AM, thus contributing to explain the T1AM phenotypic effects observed in rodents. Furthermore, T1AM influenced the expression of several genes relating to lipoprotein metabolism that provide new insights on T1AM mechanism of action, like, for example, the regulation of cholesterol homeostasis.
