Ints11 is essential for normal function of hematopoietic stem/progenitor cells [ChIP-Seq]

ABSTRACT: To investigate the role of INTS11 in normal hematopoiesis, Ints11 conditional KO mouse model was generated. We purified the Lin-cKit+ cells from bone marrow of WT and Ints11 KO mice and performed the ChIP-seq using different histone modifications to invesigate the gene regulation.

Gene-expression changes resulting from loss of the mTORC1 component Raptor in murine hematopoietic stem and progenitor cell-enriched populations (HSPC)

Project description:We investigated the role of mTORC1 in murine hematopoiesis by conditionally deleting the Raptor gene in murine hematopoietic stem cells. We observed mutliple alterations evoked by Raptor loss in hematopoiesis and profiled gene-expression alterations induced by raptor loss in Flt3-Lin-Sca1+cKit+ hematopoietic stem and progenitor enriched cell populations, 5 weeks post Raptor deletion. Flt3-Lin-Sca1+cKit+ cells were flow sorted from mice containing homozygous floxed alleles for exon 6 of the Raptor gene in the presence (MT group) or absence (WT group) of the MxCre transgene, which was induced with injections of mice with pIpC 5 weeks before cell isolation.

Project description:Deletion of Dars2 in HSPCs largely impairs spliceosome function and suppresses the protein expression of Srsf3.We performed high-coverage mRNA-seq on lin- ckit+ (LK) cells from both WT and Dars2-KO mice, respectively, to characterize genome-wide alternative splicing (AS) events affected in Dars2-KO HSPCs. Globally, 3970 AS events were significantly changed by loss of Dars2 in HSPCs, including 2431 events in skipped exons (SEs), 516 events in retained introns (RIs), 375 events in mutually exclusive exons (MXEs), 340 events in alternative 3’ splice sites (A3SSs) and 308 events in alternative 5’ splice sites (A5SSs)
