Comparison of young and senescent human endometrial stromal cells by their ability for tissue-specific decidual differentiation [RNA-Seq]

ABSTRACT: The goal of the analysis is to reveal molecular basis underlying an impairement of decidual reaction of human endometrial stromal cells (ESCs) following senescence progression. For senescence induction the model of oxidative stress induced premature senescence was applied. Young cells are non-stressed, proliferating primary ESCs of passage 8. Senescent ones — exposed to 200 µM of H2O2 for 1 hour in serum-free media and then cultured for 14 days in the complete media for senescence stabilization. Decidualization of the cells was induced by differentiation media containig 0.3 mM dibutyryl cAMP, 1 µM medroxyprogesterone 17-acetate and 10 nM β-estradiol.
