Next Generation Sequencing of Cells Derived from Directed Differentiation of Wild Type and E2A Knockout Human ESCs into Cardiomyocytes

ABSTRACT: This study aimed to compare mRNA profiles of cells derived from directed differentiation of wild type and E2A knockout human ESCs into cardiomyocytes. Using an optimized data analysis workflow, we identified 34,800 transcripts in WT and E2A -/- cardiac mesoderm cells (day3), early cardiomyocytes (day7) and later cardiomyocytes (day15). The transcripts showed 1889 differential expression genes (1099 upregulated and 790 downregulated) between the WT and E2A -/- cells, with a fold change ≥2.0 and p value <0.05. Hierarchical clustering of differentially expressed genes discovered that these genes may contribute to related phenotypes of E2A knockout cardiomyocytes.
