Single cell RNA-seq analysis on human myometrium and leiomyomas

ABSTRACT: To understand the cellular and molecular heterogeneity in the normal myometrium, we performed single-cell RNA sequencing on normal myometrium samples. We discovered 17 different cell lineages in the normal myometrium. Our work provides a single cell atlas for the normal myometrium.

Project description:The maintenance of coordinated powerful episodic contractions of the uterus is the crucial factor for normal labor. The uterine contractility is gradually enhanced with the progression of labor, which is related to the gene expression of myometrium, competing endogenous RNA (ceRNA) can also regulate the gene expression. To better understand the role of ceRNA network in labor, transcriptome sequencing was performed on the myometrium of 17 parturients at different labor duration. Furthermore, an correlated analysis was performed to identify mRNA, long non-coding RNA (lncRNA), circular RNA (circRNA), and microRNA (miRNA) which correlated with their expression levels and labor duration. Then, targeting relationships between mRNAs, lncRNAs, circRNAs and miRNAs were predicted, and the ceRNA regulatory network was established.This analysis identified 934 RNAs positively correlated with labor duration (859 mRNAs, 28 lncRNAs, 45 circRNAs, and 2 miRNAs) and 153 RNAs negatively correlated with labor duration (122 mRNAs, 28 lncRNAs, and 3 miRNAs).

Effect of Aging on the Human Myometrium at Single-Cell Resolution

Project description:The myometrial dysfunction associated with aging can prompt complications during pregnancy and labor, causing a 7.8-fold increase in maternal mortality in women over 40. Using single-cell/single-nucleus RNA sequencing and spatial transcriptomics, we constructed a cellular atlas of the aging myometrium from 186,120 cells across twenty peri- and post-menopausal women. We identified 23 myometrial cell subpopulations, including novel contractile capillary, venous capillary, immune-modulated fibroblasts, and nervous system regulatory fibroblasts. Myometrial aging leads to fewer contractile capillary cells, a reduced level of ion channel expression in smooth muscle cells, and impaired gene expression in endothelial, smooth muscle, fibroblast, perivascular, and immune cells. We observed altered myometrial cell-to-cell communication as an aging hallmark associated with the loss of 25/229 signaling pathways, including those related to angiogenesis, tissue repair, contractility, immunity, and nervous system regulation. These insights may contribute to a better understanding of the complications faced by older women during pregnancy and labor

An integrative genomic and transcriptomic analysis reveals potential targets associated with cell proliferation in uterine leiomyomas

Project description:Transcriptional profiling of uterine leiomyoma tumor samples for integrative analysis with copy number alterations data. Goal was to identify up- and/or down-expressed genes associated with genomic gains and/or losses, respectively. After, it was applied to GSEA and CONEXIC algorithm to integrate the data. Protein-protein interactions also was building and the data were validated using qRT-PCR and IHC techniques. Experiment condition, 51 uterine leiomyoma samples from patients at the secretory or proliferative menstrual cycle phases were labeled with Cy3 and adjacent normal myometrium reference samples were labeled with Cy5. Each uterine leiomyoma and adjacent normal myometrium sample were obtained from the same patient and were mixed and hybridized on a glass slide Agilent 4x44 platform.
