Project description:Astrocytes in the retrotrapezoid nucleus (RTN) stimulate breathing in response to CO2/H+, however, it is not clear how these cells detect changes in CO2/H+. Considering Kir4.1/5.1 channels are CO2/H+-sensitive and important for several astrocyte-dependent processes, we consider Kir4.1/5.1 a leading candidate CO2/H+ sensor in RTN astrocytes. To address this, we show that RTN astrocytes express Kir4.1 and Kir5.1 transcripts. We also characterized respiratory function in astrocyte-specific inducible Kir4.1 knockout mice (Kir4.1 cKO); these mice breathe normally under room air conditions but show a blunted ventilatory response to CO2, which could be partly rescued by viral mediated re-expression of Kir4.1 in RTN astrocytes. At the cellular level, astrocytes in slices from astrocyte-specific inducible Kir4.1 knockout mice are less responsive to CO2/H+ and show a diminished capacity for paracrine modulation of respiratory neurons. These results suggest Kir4.1/5.1 channels in RTN astrocytes contribute to respiratory behavior.

Project description:A parafacial region of the medulla called the retrotrapezoid nucleus (RTN) is an important respiratory control center. A group of glutamatergic neurons in this region function as respiratory chemoreceptors by regulating breathing in response to changes in tissue CO2/H+. Cellular mechanisms underlying this function involve H+-inhibition of TASK2 channels and -activation of GPR4 receptors. Evidence also suggests the RTN and greater parafacial region functions as a CO2/H+ sensing network where CO2/H+-activated and -inhibited neurons talk to each other through excitatory and inhibitory interactions. However, contributions of parafacial inhibitory neurons to control of breathing are unknown, and synaptic properties of RTN chemorecpetors have not been characterized. Here, we show the ventral parafacial region contains parvalbumin (Pvalb), cholecystokinin (CCK), neuron-derived neurotrophic factor (Ndnf) and somatostatin (SST) subtypes of interneurons including a subset strongly inhibited by CO2/H+. We also show that chemosensitive RTN neurons receive tonic inhibitory input under control conditions that is withdrawn in a CO2/H+-dependent manner, and chemogenetic inhibition of ventral parafacial inhibitory neurons increases baseline respiratory activity. These results suggest ventral parafacial inhibitory neurons are important determinants of respiratory activity under baseline conditions when the respiratory system is most prone to failure.

Project description:Purpose: compare the gene expression diffenrence between wildtype and Pten-deficient 5-HT neurons. Methods: 5-HT neurons cell body are collected from acut brain slice with patch electrodes. Pten-deficient 5-HT neurons are obtained from Pet1-Cre; Ptenflox/flox mice. Normal 5-HT neurons are obtained from littermate wildtype mice. Reverse transcription was performed using a SMARTer Ultra Low RNA Kit (Clontech, Japan). Single-cell cDNA was amplified using an Advantage 2 PCR Kit (Clontech, Japan). Purified cDNA from individual cells were sequenced using the Illumina HiSeq 2000 platform. Sequence reads were aligned to the mouse mm9 genome using TopHat. Differential expression analysis was performed using DESeq (Bioconductor) followed by Student t-test. Genes were considered differentially expressed if p<0.05. Results: The somatic compartments of individual YFP-labeled 5-HT neurons in acute brain slices were harvested for RNA sequencing. Each of the cells had 2 X 107 reads mapping back to the mouse genome when analyzed using the RUM mapping program. We found 859 genes (381 up-regulated, 478 down-regulated) were differentially expressed in Pten-deficient 5-HT neurons. Gene ontology of differentially expressed genes revealed the biological processes up-regulated and down-regulated in Pten-deficient 5-HT neurons. The absence of Pten in 5-HT neurons was associated with significantly increased expression of genes involved in the regulation of axonal extension and cell size, cell morphogenesis and cytoskeleton organization

Project description:CO2-induced vasoreactivity is an important mechanism to remove CO2 from the brain. Our previous work has shown that in brain endothelial cells Gαq and Gα11, that can replace each other and induce a common intracellular pathway, are required for normal vascular reactivity to CO2 stimulation. Additionally, we found different effects on vascular diameter dependent on the brain area. In the cortex, vessel dilate as response to CO2 stimulation, in the retrotrapezoid nucleus (RTN) of the brainstem, vessels constrict upon CO2 stimulation. To examine the potential basis for the opposing effects of CO2 on the vascular reactivity in brainstem compared to other brain areas, we prepared cortical and brainstem vessel fragments of control and Gαq/11beKO mice and performed microarrays to determine mRNA expression.

Project description:Here we performed single cell RNA sequencing of manually sorted olfactory bulb dopaminergic neurons with the goal of studying the transcriptomic basis of neuronal plasticity. Acute olfactory bulb 300 µm slices were obtained from Dat-Cre/Flox-tdTomato (B6.SJL-Slc6a3tm1.1(cre) Bkmn/J, Jax stock 006660 / B6.Cg– Gt(ROSA)26Sortm9(CAG-tdTomato)Hze, Jax stock 007909) juvenile mice. Single cell suspensions were generated using the Neural Tissue Dissociation Kit – Postnatal Neurons (Miltenyi Biotec. Cat no. 130-094-802). Dopaminergic neurons were identified by red fluorescence and manually sorted into individual wells. Analysis of the effects of the transcriptomic changes relating to neuronal plasticity triggered by sensory deprivation is ongoing.

Project description:Genome-wide gene expression was obtained in three auditory brainstem nuclei (defined below), at two different ages in mice, postnatal day (P)3 and P14. The primary aim was to identify genes which are differentially expressed between the medial nucleus of the trapezoid body (MNTB) and the superior olive (LSO), at both age groups. Tissue samples from the ventral cochlear nucleus (VCN), the medial nucleus of the trapezoid body (MNTB) and the lateral superior olive (LSO) were collected from 200 μm thick fresh transverse mouse brainstem slices prepared with a vibratome tissue slicer. Individual brainstem nuclei were dissected out with fine forceps under optical control with a binocular. For each nucleus (VCN, MNTB, and LSO), the tissue from six male C57Bl6 mice at postnatal day (P) 3, and from three male C57Bl6 mice at P14 was pooled. The RNA was isolated with the RNeasy Micro kit (Qiagen); RNA samples were amplified using a WT ovation Pico RNA amplification (Nugen, CA, USA). The cDNA was hybridized onto mouse Affymetrix microarrays 430 2.0 (Affymetrix, Santa Clara, USA).

Project description:Motor neurons are selectively vulnerable in spinal muscular atrophy (SMA). However, some brainstem motor neuron groups, including oculomotor and trochlear, which innervate the muscles around the eyes, are for unknown reasons spared. Here, we investigate the transcriptional dynamics in discrete neuronal populations in health and SMA to identify mechanisms of vulnerability and resistance. Such mechanisms could reveal targets for future gene therapy studies aimed towards preserving vulnerable motor neurons.
