Next Generation Sequencing Facilitates Quantitative Analysis of Wild Type and knock-in TrkAC lumbar dorsal roots ganglia transcriptomes in a context of arthritis induced by intra-articular injection of complete Freund's adjuvant

ABSTRACT: Our aim was to better understand the involvement of the specific intracellular signalling pathways activated by NGF through its specific tyrosine kinase type A (TrkA) receptor in the pathophysiology of rheumatoid arthritis using knock-in TrkA/C mice. This mice expresses the native extracellular part of TrkA able to bind NGF but the functional transmembrane and intracellular parts of TrkA were replaced by those of the specific receptor of neurotrophin 3 (NT3): the tyrosine kinase type C receptor (TrkC). RNA sequencing analysis was performed on lumbar (L4-L5-L6) dorsal roots ganglia of Wild Type littermates and knock-in TrkA/C mice 14 days after intraarticular injection in the ankle joint of complete freund adjuvant (CFA) or vehicle (n=4 biological samples per group).
