Transcriptome response of Aspergillus fumigatus cultures under different temperatures

ABSTRACT: Microarray analysis was used to compare gene expression of Aspergillus fumigatus under two different sporulation temperatures, 17oC and 32oC, with an emphasis on genes encoding known or putative allergens of the fungus. The objective of the study was to investigate whether allergenic potencies of A. fumigatus spores produced under different sporulation temperatures would be influenced by temperature-dependent transcriptional regulation of allergenicity genes.

Project description:Microarray analysis was used to compare gene expression of Aspergillus fumigatus under two different sporulation temperatures, 17oC and 32oC, with an emphasis on genes encoding known or putative allergens of the fungus. The objective of the study was to investigate whether allergenic potencies of A. fumigatus spores produced under different sporulation temperatures would be influenced by temperature-dependent transcriptional regulation of allergenicity genes. Non-sporulating liquid culture of Aspergillus fumigatus was harvested and divided equally onto two sets of potato dextrose agar plates, one set for incubation at 17oC, the other for incubation at 32oC. After 48 hours of incubation, RNA was harvested from both sets of sporulating cultures, reverse-transcribed into dye-coupled cDNA and hybridized onto microarrays for analysis of gene expression. For each experiment, extracted RNA from the two cultures were hybridized onto two dye-swap technical replicate arrays. A total of three separate experiments were conducted for biological triplicates, for a total of six hybridizations.

A. fumigatus spores were cultured for 48 h and then treated with artemisinin (at MIC50 concentration) or solvent control (DMSO) for 3 h to study its transcriptomic profiles.

Protein composition of Moorella thermoacetica spores formed at contrasted temperatures

Project description:Moorella thermoacetica spores are the most heat-resistant so far retrieved in food industry and we previously showed that the resistance properties of these spores to wet- heat and biocides were lower when spores were produced at low limit temperature than at optimal temperature. By electron microcopy, we observed that the ultrastructure of the spore coat differed according to the sporulation temperature, with spores produced at 55 °C mainly exhibiting lamellar inner coat tightly associated to diffuse outer coat, while spores produced at 45 °C showing an inner and outer coat separated by a less electron- dense zone. Moreover, misarranged coat structures were more frequently observed when spores were produced at low limit temperature. We analyzed the proteome of spore ob- tained at 45° and 55 °C and focused our data analysis on putative spore coat, exosporium proteins or proteins playing a role in spore resistance. Some putative spore coat proteins, such as CotSA, were only identified in spores produced at 55 °C, while some other puta- tive exosporium and coat proteins were significantly less abundant in spores produced at 45 °C. Altogether, our results suggest that sporulation temperature affects the structure and the protein composition of M. thermoacetica spores.

Project description:Foxp3+ regulatory T cells (Treg) play a central role for tolerance against self and innocuous environmental antigens. However, the role of antigen-specificity for Treg-mediated tolerance is only incompletely understood. Here we show by direct ex vivo characterization of human CD4+ T cells, that the response against innocuous airborne antigens, such as plant pollen or fungal spores, is dominated by memory-like antigen-specific Treg. Surprisingly, breakdown of tolerance in atopic donors was not accompanied by a quantitatively or qualitatively altered Treg response, but instead correlated with a striking dichotomy of Treg versus Th2 target specificity. Allergenic proteins, are selectively targeted by Th2 cells, but not Treg. Thus human Treg specific for airborne antigens maintain tolerance at mucosal sites and the failure to generate specific Treg against a subgroup of antigens provides a window of opportunity for allergy development. PBMCs from sex and age matched birch pollen allergic patients and healthy controls, were stimulated (7h) with airborne fungal (A. fumigatus) or birch pollen antigen (birch) and sorted into antigen specific conventional and regulatory T cells according to their expression of CD154+ and CD137+ on CD4+ T cells, respectively. Biotinylated surfome profiling of A. fumigatus during early development

Project description:Aspergillus fumigatus is one of the most common airborne fungi that cause invasive mycoses in immunocompromised patients and also allergic diseases in immunocompetent individuals. In both cases, the surface proteins mediate the first contact with the human immune system to evade immune responses or to induce hypersensitivity. The peptides exposed on the surface may be reasonable immunotherapy targets or may be used for diagnostic purposes. Several methods have been established to study the surface proteome (surfome) of A. fumigatus, like trypsin shaving, glucanase treatment, or formic acid extraction. Biotinylation coupled with LC-MS/MS identification of peptides is a particularly efficient method to identify the surface exposed regions of proteins that could mediate interaction with the host. After biotinylation of surface proteins during the germination process, we detected 314 surface proteins, including several well-known surface proteins (like RodA, CcpA, and DppV), allergens, heat shock proteins (Hsp), as well as some previously uncharacterized surface proteins. Using immunofluorescence microscopy, we confirmed the surface localization of several chaperone proteins. Collectively, our study generated a comprehensive data set of the A. fumigatus surfome. In addition, for many proteins it uncovers the regions that are exposed on the surface of spores or hyphae. These surface exposed regions could directly interact with host cells and may represent antigenic epitopes that induce protective immune responses.

The responses of the pulmonary pathogen Aspergillus fumigatus to the neutrophil derived oxidant N- chlorotaurine

Project description:Aspergillus fumigatus is a pulmonary pathogen to individuals who are immunocompromised (e.g. mylosuppression) and those who display defects in immune equilibrium, commonly observed in the Cystic fibrosis lung and presents itself in the form as allergic bronchopulmonary aspergillosis.32 In the latter disease A. fumigatus airborne conidia penetrate the lower airways, germinate and and grow within in thick tenacious CF mucus. A. fumigatus CF colonized individuals can become sensitized to A. fumigatus secreted allergens and eventually develop a type 2 hypersensitivity reaction to AF allergens, toxins and proteases which are readily replenished into the mucus which can directly cause cell damage and peeling at the bronco – epithelium interface. A. fumigatus also induces invasive disease which carries up to 90% mortality. The effect of neutrophil oxidant NCT on A. fumigatus was determined as neutrophils are the first line of cellular defense against A. fumigatus.

Transcriptional profiling of Aspergillus fumigatus in response to RBL-2H3 mast cells

Project description:In patients with chronic pulmonary disease colonization with the mold Aspergillus fumigatus is associated with declining pulmonary function and obstructive airway disease. One potential effector of this inflammatory response is the pulmonary mast cell. In vitro studies have demonstrated that A. fumigatus contact induces IgE-independent mast cell degranulation. Conversely, the Aspergillus secondary metabolite gliotoxin has been shown to suppress mast cell activation. These contradictory results emphasize the need for a better understanding of the interactions between A. fumigatus and mast cells. Thus, the objective of this work was to identify A. fumigatus genes that are differentially regulated upon exposure to mast cells. Transcriptional profiling experiments indicated that, in addition to genes encoding for iron acquisition systems, allergens and putative virulence factors, genes from the gliotoxin biosynthesis cluster were significantly down-regulated upon exposure to mast cells. Globally, the results from this study provide insight into the A. fumigatus response to mast cells and suggest that one mechanism by which the host may circumvent the effects of gliotoxin is via the suppression of fungal gliotoxin synthesis by mast cells.

The temporal dynamics of differential gene expression in human alveolar epithelial and endothelial cells interacting with the human pathogenic mould Aspergillus fumigatus in vitro

Project description:Aspergillus fumigatus is the most relevant pathogenic mould in immunocompromised patients. Spores of A. fumigatus are inhaled and reach the lung alveoli, where they interact with the human immune system. In this set of experiments, we co-cultured with defined morphologies of Aspergillus fumigatus (Af293) for 0h, 3h and 6h with either cells representing human alveolar epithelium (A549) or human alveolar endothelium (HPAEC). We included either monocyte-derived or myeloid dendritic cells from volunteer donor blood to the epithelial layer to examine their effect on gene expression. RNA was extracted from both cell lines at each time point and hybridised to microarrays. Microarrays contain probes for approximately 110 selected human immune-relevant genes, including cytokine and chemokine genes, their receptors and downstream innate immunity genes.
