Species comparison of liver proteomes reveals links to naked mole-rat longevity and human aging

Project description:Mammals display wide range of variation in their lifespan. Investigating the molecular networks that distinguish long- from short-lived species has proven useful to identify determinants of longevity. Here, we compared the liver of long-lived naked mole-rats (NMRs) and the phylogenetically closely related, shorter-lived, guinea pigs using an integrated omic approach. We found that NMRs livers display a unique expression pattern of mitochondrial proteins that result in distinct metabolic features of their mitochondria. For instance, we observed a generally reduced respiration rate associated with lower protein levels of respiratory chain components, particularly complex I, and increased capacity to utilize fatty acids. Interestingly, we show that the same molecular networks are affected during aging in both NMR and humans, supporting a direct link to the extraordinary longevity of both species. Finally, we identified a novel longevity pathway and validated it experimentally in the nematode C. elegans.

cest-2.2 overexpression alters lipid metabolism and extends longevity of mitochondrial mutants

Project description:Mitochondrial dysfunction can either extend or decrease Caenorhabditis elegans lifespan, depending on whether transcriptionally regulated responses can elicit durable stress adaptation to otherwise detrimental lesions. Here, we test the hypothesis that enhanced metabolic flexibility is sufficient to circumvent bioenergetic abnormalities associated with the phenotypic threshold effect, thereby transforming short-lived mitochondrial mutants into long-lived ones. We find that CEST-2.2, a carboxylesterase mainly localizes in the intestine, may stimulate the survival of mitochondrial deficient animals. We report that genetic manipulation of cest-2.2 expression has a minor lifespan impact on wild type nematodes, whereas its overexpression markedly extends the lifespan of complex I deficient gas-1(fc21) mutants. We profile the transcriptome and lipidome of cest-2.2 overexpressing animals and show that CEST-2.2 stimulates lipid metabolism and fatty acid beta-oxidation, thereby enhancing mitochondrial respiratory capacity through complex II and LET-721/ETFDH, despite the inherited genetic lesion of complex I. Together, our findings unveil a metabolic pathway that, through the tissue-specific mobilization of lipid deposits, may influence the longevity of mitochondrial mutant C. elegans.

Project description:The aim of the study is to identify genetic mechanisms of longevity and to characterize their interaction with the environment. Using the results the future health of people without familial longevity can be improved in old age. Long-lived siblings of European descent were recruited together with their offspring and the partners of the offspring. Families were recruited if at least two long-lived siblings were alive and fulfilled the age criterion of 89 years or older for males and 91 years or older for females, representing less than 0.5% of the Dutch population in 2001.

Effect of Pro-longevity drugs on Lipid profile, link between lipid signalling and longevity

Project description:We report the effect of four lifespan modifying drugs and of combinations of these drugs on lipid profile in Caenorhabditis elegans. We investigated drug-drug interactions and identified beneficial drug interactions between drugs selected to target synergistic longevity mechanisms and pathways. We employ ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS) to compare the abundance of lipid species in treated and control animals. Adult nematodes were treated with rapamycin, rifampicin, psora-4 and allantoin and combinations of these compounds and the resulting change in lipid profiles, specifically in those of triacylglycerol (TAG), phosphatidylcholine (PC) and phosphatidylethanolamine (PE) were determined. We quantified changes resulting from treatment with the drug combinations relative to untreated controls and relative to animals treated with each constituent single drugs. We further determined the dependence of changes in lipid profiles on genes know to affect lipid metabolism using strains carrying mutations in these pathways. In particular, we determined lipid profiles in genetic model of caloric restriction (eat-2), strain lacking homolog of TGFβ (daf-7) and in strain lacking the SREBP/sbp-1 transcription factor.
