A genome-wide comparison of gene expression patterns in CD4+ T cells under different T cell activating treatments

ABSTRACT: In the literature, several conditions are described that lead to T cell activation. Within this experiment we compared the influence of various T cell activating conditions on the gene expression pattern of CD4+ T cells. T cell stimulation via anti-CD3 was compared to anti-CD3/anti-CD28 as well as to anti-CD3 and IL-2 stimulation.

Project description:T-lymphocyte activation is efficiently mimicked in vitro by treatment with anti CD3 / anti CD28 antibodies. We report miR-21 induction upon CD3/CD28 stimulation of primary T-lymphocytes. In order to assess the function of miR-21 in T-lymphocytes we interfered with miR-21 function by lentiviral transduction of a miR-21 sponge construct. Differential effects of inhibition of bone morphogenic protein (BMP) signalling on T-cell activation and differentiation

Project description:Dorsomorphin is a small molecule inhibitor of type I bone morphogenic protein receptors (BMPRs). We have found that dorsomorphin affects a wide range of T cell function. In order to obtain the bigger picture of the effects of DM in T cell activation. transcriptomic analysis was performed using mouse primary CD25-CD4+ T cells with either DM (4 μM) or vehicle in the presence or absence of stimulation by anti-CD3 and -CD28 antibodies. Mouse CD4+ T cells were prepared and cultured with or without stimulation by plate bound anti-CD3 mAb and soluble anti-CD28 mAb for 1 hour. Subsequently, 4 μM DM or DMSO was added and cells were incubated for 6 hours. Total RNA was isolated using Qiagen RNeasy mini kits.

Magnetically sorted untouched CD8+CD45R0- T cells from three different donors were unstimulated or stimulated with IFNa2b or with anti-CD3/CD28 Beads alone or along with IFNa2b or IFNa5 for 48 hours. Individual mRNA samples were analyzed using HG-U133A 2.0 array gene chips. RNA-seq profiling of in vitro FRC-conditioned, IL6-conditioned or control (anti-CD3/CD28)-stimulated CD8+ T cells

Project description:To broadly assess which pathways FRCs regulate in CD8+ T cells, we sorted CD8+ T cells for RNA-seq following activation in whole splenocyte mixtures via soluble anti-CD3/CD28 for 48 hours (Stim), activated with anti-CD3/CD28 in the presence of Nos2–/– FRCs (FRC) or activated with anti-CD3/CD28 plus 100 ng/ml recombinant IL-6 (IL-6). Here we demonstrate that FRC-derived signals, including IL-6, act in concert with TCR signaling and co-stimulation to promote the expression of MYC and HIF-1-dependent glycolytic genes and vital pro-survival genes (encoding BCL-2 family members, inhibitors of apoptosis [IAPs] members and Cflar) in activated CD8+ T cells.
