Project description:Gene expression plasticity is central for macrophages? timely responses to cues from the microenvironment permitting phenotypic adaptation from pro-inflammatory (M1) to wound healing and tissue-regenerative (M2, with several subclasses). Regulatory macrophages (Mreg) are a distinct macrophage type, partially sharing some functionalities with both M1 and M2 cells. Mreg possess immunoregulatory, anti-inflammatory, and angiogenic properties, and are considered as a potential allogeneic cell therapy product to treat clinical conditions, e.g., non-healing diabetic foot ulcers. In this study we characterized clinically relevant regulatory macrophages, Mreg and Mreg_UKR, programmable cells of monocytic origin (PCMO) and comparator macrophages (M1, M2a and M0) using flow-cytometry, RT-qPCR, phagocytosis and secretome measurements, and finally RNA-Seq. We demonstrate that conventional phenotyping has a limited potential in deciphering all classes of produced cells. This limitation was ameliorated when global transcriptome characterization by RNA-Seq was employed. Using this approach we prove that macrophage manufacturing processes can result in a highly reproducible cell phenotype. At the same time, minor changes introduced in a protocol can consistently effect the phenotype of the end product as well. Additionally, we have identified a novel constellation of potential process specific biomarkers, which will not only support further clinical product development, but will lead to a better understanding of macrophage biology, differentiation and polarized activation.

Project description:Alveolar macrophages are the first line of defense against pathogens in the lungs of all mammalian species and therefore may constitute an appropriate therapeutic target cell in the treatment and prevention of opportunistic airway infections. Analysis of alveolar macrophages from several species has revealed a unique cellular phenotype and transcriptome, presumably linked to their distinct airway environment and function in host defense. The current study extends these findings to the horse. Firstly, the transcriptome of equine alveolar macrophages was compared to that of equine peritoneal macrophages. This comparison revealed the differential basal expression of 451 genes. Gene expression analysis revealed an alternative macrophage polarisation profile in alveolar macrophages and a hybrid macrophage activation profile in peritoneal macrophages, a phenomenon potentially attributable to a degree endotoxin tolerance. Secondly, the change in gene expression profile of equine AMs following lipopolysaccharide stimulation was investigated. This revealed a significant change in the expression of 240 genes, including well known upregulated inflammatory genes.

Project description:In vitro studies are crucial for our understanding of the human macrophage immune functions. However, traditional in vitro culture media poorly reflect the metabolic composition of blood, potentially affecting the outcomes of these studies. Here, we analysed the impact of a physiological medium on human induced pluripotent stem cell (iPSC)-derived macrophages (iPSDM) function. Macrophages cultured in a human plasma-like medium (HPLM) were more permissive to Mycobacterium tuberculosis (Mtb) replication and showed decreased lipid metabolism with increased metabolic polarisation. Functionally, HPLM-differentiated macrophages showed different metabolic organelle content and activity. Specifically, HPLM-cultured macrophages displayed reduced lipid droplet and peroxisome content, increased lysosomal proteolytic activity, and increased mitochondrial activity and dynamics. Inhibiting or inducing lipid droplet formation revealed that lipid droplet content is a key factor influencing macrophage permissiveness to Mtb. These findings underscore the importance of using physiologically relevant media in vitro for accurately studying human macrophage function.
