Project description:Modulation of the activity of the ubiquitin-proteasome pathway with the proteasome inhibitor (PI) is an established component of therapy for plasma cell disorders. However, resistance emerges and the mechanism is incompletely understood. We generated carfilzomib-resistant (CR) myeloma cell lines by exposing drug-naive ANBL-6, KAS-6/1, U266, and OPM-2 cells to increasing concentrations of carfilzomib and then performed gene expression profiling (GEP) to identify prominent changes compared to their vehicle-treated counterparts, followed by exploration of the mechanism(s) of proteasome inhibitor resistance.

LPS time-course in WT and Stk40 KO BMDMs

Project description:Bone marrow was harvested from Rosa26CreER; Stk40+/+ (WT; n = 3) and Rosa26CreER; Stk40loxp/loxp (Stk40 KO; n = 3) mice and differentiated for 6 days in the presence of 100 nM 4-OHT to generate WT and Stk40 KO bone-marrow derived macrophages (BMDMs). 2. On day 7 following differentiation BMDMs were treated with 100 ng x ml-1 LPS and harvested at 0 hrs, 6 hrs, 16 hrs, and 32 hrs following LPS exposure. 3. The cells were snap-frozen at the time of harvest. RNA was extracted using the Qiagen RNeasy mini kit as per manufacturer’s protocol including the on-column DNase digestion. Screening of differentially expressed genes after IMB5046 treatment

Project description:To investigate the effects of IMB5046 treatment on cells, we used the gene expression microarray to compare the difference between the IMB5046-treated cells and control cells. A431 cells were treated with 100 nM IMB5046 or 0.1% DMSO (vehicle) for 24 hr. A total of 441 genes were down-regulated and 383 genes were up-regulated by at least 2-fold between IMB5046-treated and control cells.The most obviuos changes in Gene Ontology biolocial process classification were related to cell death and immune response.
