RNA-seq of immature B cells in presence or absence of γδ T cells

ABSTRACT: Purpose: compare immature B cells in presence or not of γδ T cells in vivo and in vitro. Methods: immature B cells were sorted from spleen of control Tcrd-GDL mice and Tcrd-GDL mice treated with Diphteria toxin (DTx) for depletrion of γδ T cells after eight days and were sequenced for transcriptional (RNA-seq) and epigenetic profile (ATAC-seq). Further, immature B cells were sorted from spleen of Tcdr-H2B mice, cultured alone or in presence of γδ T cells and sequenced for transcriptional profile (RNA-seq). Results/Conclusions: In presence of γδ T cells, immature B cells upregulated genes related to unfolded protein response and mTORC1 signaling

MHC-independent αβ T cells replace epidermal and dermal γδ T cells in Tcrd-/- mice

Project description:In Tcrd-/- mice, dendritic epidermal T cells (DETC) are known to be replaced by a DETC-population expressing αβ TCR. Here, the TCR-repertoire from Tcrd-/- mouse skin T cells was compared to the one from WT mice. The analysis revealed the existence of T cells expressing MHC-independent αβ TCR and replacing the missing γδ T cells in Tcrd-/- mouse epidermis and dermis.

Single-cell RT-PCR analysis of Tgd17 progenitors

Project description:The indicated thymic progenitor population was sorted via FACS and then loaded into a Fluidigm C1 small cell capture chip for single-cell capture, lysis, reverse transcription, and preamplification. Preamplified products were analyzed on a BioMark HD with EvaGreen chemistry. Project description:RNAseq of germinal center B cells (B220+IgDlowCD95+GL7+) with different Blimp-1-GFP reporter level in the presence of TfH cells or 72 hr after their depletion. Sorted germinal center populations were defined by their expression of Blimp-1-GFP reporter and the cell surface marker CD138. Analysed mice were bone marrow chimeras generated with a bone marrow mixture from CD4-cre+ Rosa26-loxP-stp-loxP-DTR Blimp-1GFP/+ (10%) and Rag1-/- OTII (90%) mice. In these mice TfH cells can be depleted by the injection of human diphteria toxin (DT).

Project description:We generated mice with a mutation at threonine 58 of MYC in the mouse germline (T58A mutant). These mice ultimately develop AML or B cell lymphomas. We profiled changes in gene expression and genomic occupation of MYC in single cells of sorted, immature bone marrow progenitor cells, mature (spleen derived) and immature (marrow derived) B cells. B cells were stimulated with LPS and IL7, respectively.
