RNASeq, multiome, and genomic profiling of hematopoietic progenitors and B cells from mice with a point mutation in MYC [CUT&RUN]

ABSTRACT: We generated mice with a mutation at threonine 58 of MYC in the mouse germline (T58A mutant). These mice ultimately develop AML or B cell lymphomas. We profiled changes in gene expression and genomic occupation of MYC in single cells of sorted, immature bone marrow progenitor cells, mature (spleen derived) and immature (marrow derived) B cells. B cells were stimulated with LPS and IL7, respectively.

Project description:We sought to examine the mechanism through which phospho-mutants can contribute to the transformation of MCF10A acini. To investigate this, we examined the RNA abundance of Myc and Myc phospho-mutants (T58A, S71A/S81A, and Myc-4A) against a GFP control. Three independent biological replicates of the five samples (Myc, T58A, S71A/S81A, Myc-4A and GFP control) were isolated on day 4 and processed on Affymetrix HG-U133 Plus2 chips.

Project description:Hematopoiesis in adult mammals involves cognate interactions between developing hematopoietic cells and bone marrow stromal cell niches. SCF and CXCL12 play key roles in the maintenance of HSC, while early B cell differentiation requires CXCL12 and IL7. In this study, we characterized mouse BM stromal cells expressing IL7 by performing a transcriptomic analysis. We found that SCF, CXCL12 and IL7 were co-expressed by a unique peri-sinusoidal stromal cell subset.

Peri-sinusoidal stromal cells form a unique multispecific niche for hematopoietic stem cells and early B cells

Project description:In the bone marrow, CXCL12 and IL7 are essential for B cell differentiation whereas hematopoietic stem cells (HSC) maintenance requires SCF and CXCL12. Previous studies, including ours, indicate that peri-sinusoidal stromal (PSS) cells are heterogeneous and that part of them express and are the main source of IL7. Our single cell qPCR and transcriptomic analysis demonstrate that SCF, CXCL12 and IL7 are co-expressed by a unique peri-sinusoidal stromal cell subset.

PIM447 treatment in T-ALL PDX splenocytes

Project description:A subset of T-ALL/T-LBL patients is characterized by overexpression of the oncogenic Serine/Threonine kinase PIM1. Mechanisms for PIM1 overexpression are translocations involving the TCRb and PIM1 loci, JAK/STAT activating mutations and responsiveness to cytokinens in the tumor microenvironment, such as IL7. In this study, we generated a TLX3+ JAKwt/STATwt IL7 responsive T-ALL PDX model using NSG mice. Splenocytes were ex vivo treated with the PIM inhibitor PIM447 and QuantSeq 3’ mRNA sequencing was performed, in order to elucidate the transcriptional effects and molecular mechanisms by which PIM447 induces apoptosis in T-ALL PDX cells.
