Imetelstat is effective in Acute Myeloid Leukemia (AML) patient xenografts.

ABSTRACT: Imetelstat is a covalently lipidated 13-mer thiophosphoramidate oligonucleotide that competitively inhibits telomerase activity. Here, we show that imetelstat effectively diminishes AML burden and prolongs survival in a comprehensive, randomized Phase II-like preclinical trial in AML patient-derived xenografts (PDX).

Project description:Analysis of gene expression profiles of human AML patient samples. The hypothesis tested was that AML patient samples showing a higher sensitivity to subsequent telomerase inhibition would already harbour a telomerase deficiency gene expression signature before treatment that may be useful to predict therapeutic efficacy. The results provide a clear separation of AML patient samples based on a telomerase deficiency gene signature that is correlated with subsequent sensitivity to pharmacologic telomerase inhibition assessed in a xenograft transplantation system in vivo.

Synergistic Anti-Leukemic Effect Of Trc105 And Decitabine In AML Xenografts

Project description:Acute myeloid leukemia (AML) is an aggressive hematologic malignancy, often characterized by poor prognosis following standard induction therapy. The DNA methyltransferase inhibitor decitabine (DAC) is an alternative to intensive chemotherapy in elderly and relapsed/refractory AML patients. Despite some efficacy, DAC monotherapy does not promote complete remission and overall survival rates are very poor. Therefore, novel therapeutic regimens for AML are warranted. CD105 (endoglin), a receptor for the TGF-β superfamily, is expressed in various hematopoietic malignancies, including AML in which high expression correlates with poor prognosis. We have recently demonstrated that targeting CD105+ AML blasts with the TRC105 monoclonal antibody abrogates leukemogenesis in vivo. Based on these encouraging findings, here we investigated whether TRC105 in combination with DAC could represent a novel therapeutic option to treat relapsed/refractory AML. Treatment of leukemic mice with DAC in combination with TRC105 resulted in superior survival and significant reduction of leukemia burden in the bone marrow (BM) of AML xenografts compared to DAC monotherapy. To determine if TRC105 anti-leukemogenic effect is mediated by NK cells, we performed in vitro and in vivo studies. In the latter, we tested TRC105 in the context of NK depletion by injecting mice with an antibody to CD122. TRC105 treatment resulted in comparable reduction of leukemia burden in xenografts regardless of NK depletion, demonstrating that the anti-leukemogenic effect of TRC105 in AML is independent of NK cells. Altogether, our findings provide proof-of-principle for the clinical evaluation of TRC105 in combination with DAC in AML patients.

Temporal Control of Mammalian Cortical Neurogenesis by m6A Methylation

Project description:N6-methyladenosine (m6A), installed by the Mettl3/Mettl14 methyltransferase complex, is the most prevalent internal mRNA modification. Whether m6A regulates mammalian brain development is unknown. Here we show that Mettl14 deletion in the embryonic mouse brain diminishes m6A levels, prolongs cell cycle of radial glia cells, and extends cortical neurogenesis into postnatal stages. Mettl3 knockdown also prolongs neural progenitor cell cycle and promotes radial glia cell maintenance. m6A-sequencing of the embryonic mouse cortex reveals enrichment of mRNAs related to transcription factors, cell cycle and neuron differentiation, and Mettl14 deletion attenuates their decay. Notably, Mettl14-/- radial glia cells precociously express neuronal proteins. Further analysis uncovers previously unappreciated transcriptional pre-patterning in cortical neural stem cells. Comparison of m6A-mRNA landscapes between mouse and human cortical neural progenitors identifies human-specific tagging of transcripts related to epigenetic regulation and brain disorder risk genes. Our study reveals an epitranscriptomic mechanism in heightened transcriptional coordination during mammalian cortical neurogenesis.

Effective Menin inhibitor-based combinations against AML with MLL rearrangement or NPM1 mutation (NPM1c)

Project description:Treatment with Menin inhibitor (MI) disrupts interaction between Menin and MLL1 or MLL1-fusion protein (FP), inhibits HOXA9/MEIS1, induces differentiation and loss of survival of AML harboring MLL1 re-arrangement (r) and FP, or expressing mutant (mt)-NPM1. Following MI treatment, although clinical responses are common, majority of patients with AML with MLLr or mt-NPM1 succumb to their disease. Pre-clinical studies presented here demonstrate that genetic knockout or degradation of Menin, or treatment with the MI SNDX-50469 reduces MLL1/MLL1-FP targets, associated with MI-induced differentiation and loss of viability. MI treatment also attenuates BCL2 and CDK6 levels. Co-treatment with SNDX-50469 and BCL2 inhibitor (venetoclax), or CDK6 inhibitor (abemaciclib) induces synergistic lethality in cell lines and patient-derived AML cells harboring MLL1r or mtNPM1. Combined therapy with SNDX-5613 and venetoclax exerts superior in vivo efficacy in cell line or PD AML cell xenografts harboring MLL1r or mt-NPM1. Synergy with the MI-based combinations is preserved against MLLr AML cells expressing FLT3 mutation, also CRISPR edited to introduce mtTP53. These findings highlight the promise of clinically testing these MI-based combinations against AML harboring MLL1r or mtNPM1.
