CD16+ CD163+ monocytes traffic to sites of inflammation in NEC

ABSTRACT: Using suspension (CyTOF) and imaging (IMC) mass cytometry coupled with single cell RNA sequencing (scRNAseq) on intestinal mucosa, we demonstrated severe inflammation in patients with NEC. scRNAseq revealed that the CD163+CD16+ monocytes/Mf transcribe a large number of inflammatory genes including TREM1, IL1a and IL1b, IL8 and calprotectin. Gene set enrichment analysis identified pathways involved in chemotaxis and migration to be enriched in the clusters containing CD163+CD16+ monocytes/Mf, confirming our observations that they traffic from the periphery.
