Project description:Purpose: Hepatic stellate cells (HSCs) plays an important role in liver fibrosis. Recombinant human Cytoglobin (rhCYGB) can inhibit the activation of HSCs. The goal of this study is to investigate the rhCYGB targeted signalling pathways regulating the activation of HSCs. Method: mRNA profiles of HHSteCs control and HHSteCs treated with rhCYGB (80 µg/ml, for 48 hour) were generated by deep sequencing. Results: The RNA-seq data confirmed that extracellular matrix-encoding genes and fibrosis-related genes were down-regulated by the His-CYGB treatment. The top of up-regulated genes following His-CYGB treatment were interferon (IFN)-stimulated genes (ISGs), implying IFN signaling in His-CYGB treatment.

However, the underlying mechanisms remain unclear. Project description:We and others have shown that S100P is highly upregulated during the progression of pancreatic cancer. We used microarrays to look at the target genes regulated by S100P in the pancreatic cancer cell line Panc1. Keywords: Gene overexpression We generated stable cell lines by introducing control vector pcDNA3.1/V5-His or S100P-overexpressing vector pcDNA3.1/S100P-V5-His into the pancreatic cancer cell line Panc1, single cell clones were then isolated. RNA was extracted and hybridized on Affymetrix microarrays. We looked for new target genes regulated by S100P.

Cytoglobin overexpression-dependent transcriptomic changes in A375 melanoma cells

Project description:Cytoglobin (CYGB) is a member of the oxygen-binding globin superfamily. In this study, we comprehensively explored the CYGB-dependent transcriptome in A375 melanoma cells overexpressing CYGB. Our findings reveal that CYGB overexpression positively enriches cancer-associated pathways, including the mTORC1 and AKT/mTOR signaling pathways, which are frequently overactivated in tumors. Moreover, several cancer-associated pathways, such as epithelial-mesenchymal transition (EMT) mediated by CSPG4, were downregulated upon CYGB overexpression. Intriguingly, our results indicate that CYGB overexpression leads to a reduction in the inflammatory state of melanoma cells. This anti-inflammatory potential is exemplified by the downregulation of key inflammasome-associated genes, including NLRP1, CASP1, and CD74, which play pivotal roles in cytokine regulation and inflammasome activation. Consistent with established globin functions, CYGB appears to play a crucial role in redox homeostasis. Furthermore, our study highlights CYGB's involvement in DNA repair mechanisms and its regulation of NOX4, reinforcing its versatility in safeguarding genomic integrity. Collectively, our data illuminate the diverse functions of CYGB in melanoma cells, pointing to its roles in cellular protection against oxidative stress, inflammation, and cancer-associated pathways. These findings pave the way for further research into the physiological role of CYGB and its potential as a therapeutic target in melanoma.

Circular RNA expression signature of low-glucose-treated MIA PaCa-2 cells

Meanwhile, circular RNAs, a subset of noncoding RNAs, plays an important role in glucose metabolism and cancer progression. Analysis of low-glucose-treated and normal-glucose-treated MIA PaCa-2 cells from 6 samples (3 samples each group) was conducted. Results indicate insight into molecular signature of the pathogenesis of PDAC in abberant glucose metabolism.

Effects of PGRMC1 phosphorylation status on MIA PaCa-2 cells

Project description:Progesterone Receptor Membrane Component 1 (PGRMC1) is expressed in many cancer cells, where it is associated with detrimental patient outcomes. Multiple different functions and cellular locations have been attributed to PGRMC1 in a variety of contexts, however the mechanisms underlying PGRMC1 biology remain obscure. The protein contains several phosphorylated residues including tyrosines which were acquired early in animal evolution and could be involved with animal cell differentiation mechanisms. Here we demonstrate that mutagenic manipulation of PGRMC1phosphorylation status in MIA PaCa-2 (MP) pancreatic cells exerts broad pleiotropic effects, influencing cell plasticity and tumorigenicity, as assayed by cell biological and proteomics measurements. Relative to MP cells over-expressing hemagglutinin (HA)-tagged wild-type PGRMC1- HA (WT), cells expressing a PGRMC1-HA-S57A/S181A double mutant (DM) exhibited reduced levels of proteins involved in energy metabolism and mitochondrial function. This was associated with Rho-kinase inhibitor (ROCKI)-sensitive changes including altered cell shape, motility, increased PI3K/Akt and JNK activity, and fragmented mitochondrial morphology. An S57A/Y180F/S181A triple mutant (TM) reduced PI3K/Akt and JNK activation, indicating involvement of Y180. Both TM cells and Y180F single mutant cells exhibited attenuated mouse xenograft tumor growth. Tyrosine 180 phosphorylation status of PGRMC1 exerts dramatic influence over cancer cell biology.
