Transcriptional profiling of Regulatory T cells isolated from ApoE-/- and wild type mice

ABSTRACT: Purpose: The goals of this study were to identify preferential gene expression signatures that are unique to Tregs in the development of atherosclerosis. Methods and results: ApoE-/- and widetype mice were fed with high fat diet for 12 weeks.CD4+Foxp3+ Tregs from spleen were purified by cell sorting (using the Treg GFP reporter mouse line Foxp3/GFP) to generate mRNA transcription profiles. Transcriptional profiling revealed a unique Treg signatures in ApoE-/- mice relative to wildtype mice. Conclusion: Our study represents the first detailed analysis of splenic Treg transcriptome in atherosclerotic mice.
