Project description:In this study the transcriptional response of an ExPEC E. coli strain (CFT073) to human serum was investigated. In response, CFT073 up-regulated expression of iron and manganese acquisition systems and induced expression of iron regulated genes. High osmolarity of serum induced the osmotic shock response genes, promoting uptake of osmoprotectants by CFT073. Resistance of CFT073 to the bactericidal properties of serum involved increased expression of envelope stress regulators including CpxR, ?E and RcsB. Many of the up-regulated genes induced by active serum were regulated by the Rcs two component system. This system is triggered by envelope stress such as changes to cell wall integrity. RcsB-mediated serum resistance was conferred through induction of the exopolysaccharide colanic acid. Production of this exopolysaccharide may be protective while cell wall damage caused by serum components is repaired. Experimental Design: Two experiments are reported: 1) . The transcriptome of E. coli CFT073 exposed to LB supplemented with 50 % normal human serum was compared to that of bacteria grown in LB alone for 45 min, and 2) The transcriptome of CFT073 in response to normal healthy serum and heat inactivated serum (which has no bactericidal activity). Project description:In Escherichia coli, the Twin-arginine (Tat) secretion system is one of the main routes of the protein export to the periplasm. The Tat pathway secretes a set of proteins with important physiological functions. In our study, we investigated the influence of the deactivation of the Tat pathway on the E. coli cells. We applied a comprehensive and comparative proteomic analysis of the E. coli wild type and tat mutant. This dataset provides mass spectrometry based details on the abundances of proteins in cytoplasmic, periplasmic and membrane fractions. We observed that a tat deletion increases abundances of proteins involved in protein folding, degradation, responses to heat, oxidation, osmolarity, and cold. Moreover, the impairment of E. coli outer membrane resulted in the activation of proteins responsible for cell wall biogenesis. The tat deletion negatively affects the synthesis of iron transporters and imbalances its homeostasis in the cell.

Project description:The twin-arginine translocation (Tat) system is involved in a variety of important bacterial physiological processes. Conserved among bacteria and crucial for virulence, the Tat system is deemed as a promising anti-microbial drug target. However, the mechanism of how the Tat system functions in bacterial pathogenesis has not been fully understood. In this study, we showed that the Tat system was critical for the virulence of an extra-intestinal pathogenic E. coli (ExPEC) strain PCN033. A total of 20 Tat-related mutant strains were constructed, and competitive infection assays were performed to evaluate the relative virulence of these mutants. The results demonstrated that several Tat substrate mutants, including the ΔsufI, ΔamiAΔamiC double mutant as well as each single mutant, ΔyahJ, ΔcueO, and ΔnapG, were significantly outcompeted by the WT strain, among which the ΔsufI and ΔamiAΔamiC strains showed the lowest competitive index (CI) value. Results of individual mouse infection assay, in vitro cell adhesion assay, whole blood bactericidal assay, and serum bactericidal assay further confirmed the virulence attenuation phenotype of the ΔsufI and ΔamiAΔamiC strains. Moreover, the two mutants displayed chained morphology in the log phase resembling the Δtat and were defective in stress response. Our results suggest that the Tat system and its dependent cell division proteins SufI, AmiA, and AmiC play critical roles during ExPEC pathogenesis.

Despite of its importance, pathogenicity mechanisms of APEC strains remain not completelly elucidated and available vaccines are not fully effectives. In order to better understand which genes could be related to pathogenicity in different APEC isolated, a microarray analyses of two APEC strains representing: Swollen Head Syndrome and Omphalitis was carried out.
