Project description:Circulating human red blood cells (RBCs) consist of mature erythrocytes, and immature reticulocytes. As being anucleated cells, RBCs lack typical, abundant transcriptomes but are known to contain low amounts of diverse long transcripts and microRNAs. The exact role and importance of these RNAs is, however, lacking. To study this further, we have explored the RNA content of RBCs as well as extracellular vesicles of RBCs using next-generation sequencing (NGS). Furthermore, to understand the dynamics of the RBC transcriptome, we performed single cell RNA sequencing on RBCs both from fresh blood and blood unit. Analysis of the single cell transcriptomes revealed that while the majority of the cells don’t have detectable RNA, the remaining, approximately 10% of the cells fall into three sub-populations based on their RNA content. Overall decrease in the RNA quantity over the populations is observed. Qualitative changes include the differences in the hemoglobin (Hb) content of the cells, and the changes in the expression of ribosomal and mitochondrial genes. A previously unknown transcript of a long non-coding RNA, MALAT1 (Metastasis Associated Lung Adenocarcinoma Transcript 1) is the most enriched marker gene for one subpopulation of RBCs, suggesting a role for MALAT1 in reticulocyte maturation. Enriched pathways in RBC transcriptome, such as autophagy, reflect the processes required for reticulocyte maturation to erythrocytes. RBC transcriptome is transferred to extracellular vesicles (EVs), suggesting the vesiculation as the mechanism to remove the cellular contents at the final stages of erythrocyte maturation.

Project description:In this project we explore the cellular heterogeneity of a mouse model of heart failure with preserved ejection fraction (HFpEF) involving a two-hit model of feeding a high fat diet (HFD) along with L-NAME administration. Healthy adult male mice (C57BL/6J inbred) were fed either a normal chow diet or HFD/L-NAME for 10 weeks or 15 weeks before performing sequencing experiments. Both cardiomyocytes (CMs) and total interstitial population (TIP) were captured using a protocol to jointly capture and sequence single-nuclei (for cardiomyocytes) and single-cells (for TIP) using the 10x Genomics Chromium system.

Project description:Red blood cells (RBC) can act as carriers for therapeutic agents and given their biocompatibility and long lifespan in the circulation, they can substantially improve the safety, pharmacokinetics, and pharmacodynamics of many drugs. Maintaining RBC integrity and lifespan is important for the efficacy of RBC as drug carrier and can be a complex challenge of drug encapsulation. We investigated the impact of drug encapsulation by hypotonic dialysis on RBC physiology and integrity. Several parameters were compared between processed RBC loaded with L-asparaginase (“eryaspase�), processed RBC without drug and non-processed RBC. Processed RBC were less hydrated and displayed a reduction of intracellular content, relative to non-processed RBC. This reduction of intracellular content changed the RBC metabolomic profile, as indicated by the activation of the pentose phosphate pathway, but no impact on the global RBC proteomic profile was observed. We found that the encapsulation process caused moderate morphological changes and was accompanied by an increase of microparticles. Despite a decrease in their deformability, processed RBC were not mechanically retained in a spleen-mimicking device. Moreover, processed RBC had increased surface-to-volume ratio and osmotic resistance. Finally, the half-life of processed RBC was not significantly affected in a mouse model and our previous phase 1 clinical study showed that encapsulation of asparaginase in RBC prolonged its in vivo half-life compared to free form. Overall, our study demonstrated that encapsulation by hypotonic dialysis may affect certain several characteristics of RBC, but does not significantly impact on the in vivo longevity of RBC or their drug carrier function.

Project description:<p>Circulating tumor cells (CTCs) are recognized as direct seeds of metastasis. However, CTC count may not be the 'best' indicator of metastatic risk because their heterogeneity is generally neglected. In this study, we develop a molecular typing system to predict colorectal cancer metastasis potential based on the metabolic fingerprints of single CTCs. After identification of the metabolites potentially related to metastasis using mass spectrometry-based untargeted metabolomics, setup of a home-built single-cell quantitative mass spectrometric platform for target metabolite analysis in individual CTCs and use of a machine learning method composed of non-negative matrix factorization and logistic regression, CTCs are divided into two subgroups, C1 and C2, based on a 4-metabolite fingerprint. Both <em>in vitro</em> and <em>in vivo</em> experiments demonstrate that CTC count in C2 subgroup is closely associated with metastasis incidence. This is an interesting report on the presence of a specific population of CTCs with distinct metastatic potential at the single-cell metabolite level. </p>

Project description:Red blood cell (RBC) invasion by malaria merozoites involves formation of a parasitophorous vacuole into which the parasite moves. The vacuole membrane then seals and pinches off behind the parasite through an unknown mechanism, enclosing it within the RBC. During invasion, several merozoite surface proteins are shed by a membrane-bound protease called SUB2. Here we show that genetic depletion of SUB2 abolishes shedding of a range of parasite proteins, identifying previously unrecognized SUB2 substrates. Interaction of SUB2-null merozoites with RBCs leads to either successful entry but developmental arrest, or abortive invasion with rapid RBC lysis. Selective failure to shed the most abundant SUB2 substrate, MSP1, reduces intracellular replication, whilst conditional ablation of the substrate AMA1 produces host RBC lysis. We conclude that SUB2 activity is critical for the correct functioning of merozoite surface protein substrates and for host RBC membrane sealing following parasite internalisation.
