RNAseq of two mouse LUSC cell lines derived from NTCU-induced model

ABSTRACT: We report RNAseq data form two independent mouse syngeneic LUSC cell lines UN-SCC679 and UN-SCC680 Carcinogenesis and cell line generation: LUSC tumors were induced by N-nitroso-tris-chloroethylurea (NTCU) (Toronto Research Chemicals) treatment applying 0.04 M NTCU by skin painting twice a week for 20 weeks to 8-week-old A/J mice. Lungs of euthanized mice were excised and tumor cell lines UN-SCC679 and UN-SCC680 were derived and cultured for 25 passes to ensure they were immortalized, subcutaneously injected into 6-week-old female Rag2-/-IL2Rg-/- immunodeficient mice flanks subcutaneously and finally engrafted in syngeneic 8-week-old A/J mice. These cell lines were cultured in RPMI 1640 supplemented with 10% Fetalclone (Thermo Fisher Scientific) and 100 U/mL penicillin-100 µg/mL streptomycin (Thermo Fisher Scientific). All cells were grown in a humidified incubator containing 5% CO2 at 37°C. Cell lines were routinely tested for mycoplasma.
