Tomato xylem proteome composition following infection by Fusarium oxysporum

Project description:The influence of during colonization by Fusarium oxysporum f. sp. Lycopersici secreted effector proteins on the proteome of the xylem sap of tomato plants was investigated using a label-free quantitative proteomics approach. A comparison was made between plants inoculated with either a mock control, a non-effector knockout control, Fusarium oxysporum Fol007 wildtype and four Fol007 single effector protein knockout strains. Specific effects on the relative abundance of certain proteins of the xylem sap occurred for the different knockout strains next to a core set of 24 differentially accumulated proteins which may provide insights into the mechanisms of promoting infection for each of the tested effector proteins.

Fusarium oxysporum f.sp. radicis-lycopersici induces distinct transcriptome reprogramming in resistant and susceptible isogenic tomato lines

Project description:Background: Fusarium oxysporum f.sp. radicis-lycopersici (FORL) is one of the most destructive necrotrophic pathogen of tomato, causing important field and greenhouse yield losses. Despite the relevant economic impact, little is known about the molecular mechanisms regulating the Fusarium oxysporum f.sp. radicis-lycopersici resistance in tomato. Results: A transcriptomic experiment was carried out in order to investigate major mechanisms of FORL response in resistant and susceptible isogenic tomato lines. The microarray analysis at 15 DPI reveals a distinct gene expression pattern between the two genotypes in the inoculated vs not inoculated conditions, furthermore confirmed by a q-PCR experiment. A model of plant response for both compatible and incompatible reaction was proposed. In particular, in the incompatible interaction an activation of defense genes related to secondary metabolites production and tryptophan metabolism was observed. Moreover, the cell osmotic potential maintenance after the FORL roots challenging was mediated by a dehydration induced protein. As for the compatible interaction the activation of an oxidative burst mediated by Peroxidases and a Cytochrome monooxigenase, conducedthe cells to degeneration and necrosis. Conclusions: Our work allowed a comprehensive understanding of the molecular basis of the tomato-FORL interaction. Result obtained emphasizes a different transcriptional reaction between the resistant and the susceptible genotype to FORL challenging. Our findings could lead to the improvement of strategies of this disease control.

Project description:The gram- positive bacterial pathogen Clavibacter michiganensis subsp. michiganensis (Cmm) causes huge economic losses by infecting tomato plants worldwide. Cmm can be spread by contaminated seeds and transplants, penetrating the plant through natural openings or wounds and is transferred through the plant xylem. While in recent years significant progress has been made to elucidate plant responses to pathogenic gram-negative bacteria by gene expression studies, the molecular mechanisms that lead to disease symptoms caused by gram-positive bacteria like Cmm remain elusive. An indigenous virulent Cmm strain isolated from a farm crop of Pomodoro tomatoes in southern Greece was used for the infection of EKSTASIS F1 hybrid tomato seedlings. Here, we present the results of a deep RNA- sequencing (RNA-seq) analysis performed to characterize the dynamic expression profile of tomato genes upon Cmm infection.

Genetic Dissection of Photosynthetic Organs Contribution to Seed Oil Accumulation in Arabidopsis seeds

Project description:We report the application of RNA-seq technology for highthroughput profiling of photosynthetic and non-photosynthetic seeds of Arabidopsis chlorophyll synthase mutant seeds. By generating over 21 GB of sequence data from these seeds, we showed that genes involved in oil accumulation in non-photosynthetic seeds were significantly induced compared to photosynthtic seeds. Additionally we found that genes involved in the plastidal oxidative pentos phosphate pathway were significantly upregulated in the non-photosynthetic seed opposite to photosynthetic seeds. Overall our RNA-seq analysis revealled the genes and pathway interaction underpinining oil accumulation in non-photosynthetic seeds.

Project description:Fusarium head blight (FHB) is a major disease of cereal crops caused by the fungus Fusarium graminearum (Fg). FHB affects the flowering heads (or spikes) and developing seeds. This study compare the gene expression profile in wheat spikelets (spk 2) inoculated with either water (mock treatment) or a pathogenic strain of Fusarium graminearum (WT); spikelets 2 were inoculated 24 hrs after a neighbour spikelet (spk 0) was treated with either water or F. graminerum mutant strain Tri6Δ or NoxABΔ. Spikelets 2 were sampled 8 and 24 hrs after the second treatment.

Identification and functional characterization of calcineurin substrates in tomato Fusarium wilt fungus

Project description:Tomato is a worldwide economic crop, and Fusarium wilt is one of the important tomato diseases which is caused by Fusarium oxysporum f. sp. lycopersici (Fol). The typical symptoms are yellowing and wilting on leaves and browning vascular tissues. Subsequently, it may cause tomato to die or no fruit production. Calcineurin is a heterodimeric phosphatase which can be activated by Ca2+/calmodulin and can dephosphorylate downstream substrates to regulate stress survival, growth, sexual differentiation, and virulence. Calcineurin substrates have been studied in Saccharomyces cerevisiae by phosphoproteomics and quantitative mass spectrometry. To date, there is no research related to proteome analysis of calcineurin downstream substrates in plant pathogens. In our previous study, it revealed that calcineurin is responsible for hyphal growth, conidia formation, chlamydospore formation, and virulence in Fol. In our phosphoproteomics analysis, we compared the protein abundance of wild -type and Δcna1 strains with calcium or non-calcium induced condition. Next, we used Scanprosite and Uniport to predict conserved docking and dephosphorylated sites within those proteins by the abundance more than 2-fold distribution. Finally, we obtained 59 putative calcineurin substrates. According to GO enrichment analysis, these substrates included transcription factors, transmembrane transporters, protein serine/threonine kinase activator, and some uncharacterized proteins. We propose that these putative substrates may play important roles to control physiological function and virulence in Fol.
