Liver transcriptome analysis of mice with ASirs2 suppression

ABSTRACT: Purpose: We show that the Irs2 locus, a critical regulator of insulin actions, encodes an antisense transcript ASIrs2, whose expression increased after refeeding in liver, reciprocally to that of Irs2. In order to elucidate the functions of ASIrs2, the liver transcriptome was analyzed under ASIrs2 suppression in wild type mice after refeeding. Methods: C57BL/6 mice were injected with AAV encoding shRNA for ASIrs2 via the via tail vein at 8 weeks of age, and the mice were sacrificed for organ collection after overnight fasting and subsequently 6 hours of refeeding on the 27th or 28th day after AAV injection.
