ABSTRACT: We compared transcriptome profiling (bulk RNA-seq) of hepatic stellate cells isolated from mouse livers by perfusion method using collagenase at 37 degrees or 6 degrees and evaluated the heat-stressed gene expression at 37 degrees.

Project description:In this data set we investigate dynamic differences in the transcriptome between 37 and 40 degrees C after treatment with TNFalpha. In the continuous temperature experiments, cells were grown at 37 degrees C and were then left at 37 degrees C, or transferred to 40 degrees C, for 1hour after which they (at time 0) were stimulated with 10 ng/ml of TNFÎ± . Measurements were taken at 0, 30, 130, 230 and 430 minutes. In the 'switch' experiment, cells were grown the same way but the temperature of the incubator was turned up to 40 degrees C after 200 minutes. Measurements were taken at 230 and 430 minutes.

Project description:This experiment investigates the effect of mild cold-shock (32 degrees C, 24 hours) on the translatome of human HEK293 cells. After cold-shock, cells were treated with cycloheximide to block elongation, then sucrose density gradients were used to separate post-nuclear extracts into polysome and subpolysome fractions. The pooled polysomal fraction was labelled with Cy5, the pooled subpolysomal fraction was labelled with Cy3, and both were hybridised to the same array.

Project description:This data was utilized to assess the utility of perfusion culture in cultivating spheroids of sarcoma cells. The differences between spheroids cultured with perfusion and without perfusion were analyzed through mass spectrometry. The spheroids were fabricated from NCC-UPS4-C1 cell line derived from a patient with undifferentiated pleomorphic sarcoma.
