RNA-Seq analysis of PuCH35 overexpression line and wildtype Populus ussuriensis root tips in response to PEG stress

ABSTRACT: One week-old in-vitro plants of PuCH35 overexpression (PuC3H35-OE) lines and wildtype (WT) Populus ussuriensis were transfered to 1/2 MS medium supplemented with or without 6% PEG6000 for 7 days, and the root tips (0.5 cm long) were harvested. Samples were collected from three biological replicates (25 in-vitro plants each).

Project description:Drought is one of the major environmental problem in terms of limiting the survival of plant. Roots are the first plant organ to sense drought stress, so it is important to understanding of the molecular machanisms of drought stress responses in the roots. Here we aim to characterized the gene expression profile in Populus ussuriensis roots at 0, 6, 12, 24, 48 and 120 h after the start of PEG-induced drought stress. A total of 2 μg RNA per sample was used to generate sequencing libraries using the NEBNext® Ultra™ RNA Library Prep Kit for Illumina® (#E7530L, NEB, USA). Libraries were sequenced in 150 bp paired-end mode, using an Illumina HiSeq X Ten platform. Our results provide a global view of gene expression profile that contributes to drought resistance in Populus ussuriensis, and meaningful information for genetic engineering research in the future.

Project description:Gene expression response of Populus plants subjected to leaf wounding was analyzed using the Affymetrix poplar genome microarrays. Experiment Overall Design: Populus fremontii x angustifolia genotypes were grown hydroponically.Â Wounding treatment was applied by pinching leaves at Leaf Plastchron Index (LPI) 8-12 with pliers at a rate of 10 1-inch wounds per leaf.Â Root tips (terminal 1 cm) and mature leaves (LPI 5) were sampled 90 hours after the treatment. Two biological replicates were obtained for each condition.
