An intersectional analysis of LncRNAs and mRNAs reveals the potential therapeutic targets of Bi Zhong Xiao Decoction in collagen-induced arthritis rats

ABSTRACT: Rheumatoid arthritis (RA) is a chronic progressive autoimmune disease of unknown etiology. Both clinical and experimental studies have revealed the effects of the traditional Chinese formula Bi Zhong Xiao Decoction (BZXD) in treating RA. However, the multifaceted mechanisms of BZXD in RA treatment are not comprehensively understood. In this study, we aimed to explore therapeutic targets of BZXD by using lncRNA and mRNA transcriptomics.

Quantitative Proteomics Revealed the Changes of Pharmacodynamic Network of Bugu Shengsui Decoction Promoting Osteoblasts Proliferation

Project description:Insufficient osteogenesis is greatly essential to osteoporosis. Bugu Shengsui Decoction, a compound formula for osteoporosis, has significant clinical effects in the treatment of osteoporosis. Yet the detailed mechanisms are unclear. In this study, we first evaluated the pharmacological effects of Bugu Shengsui Decoction on bone metabolism, bone mineral density, bone morphology and biomechanics in ovariectomized rats. It also clarified the promoting effect of Bugu Shengsui Decoction on the proliferation and differentiation of osteoblasts. Based on the above results, we conducted quantitative proteomics research and a series of validation experiments, the results showed that PI3K-AKT signaling pathway and targets, such as Fn1, Col1a1, Col1a2, Col6a1, Col6a2, Col6a3, Rac1, Hsp90ab1, Hsp90b1, Ywhaz and Gnb2, were most relevant to the anti-osteoporosis efficacy of BGSSD. Summarily, our discoveries certify that Bugu Shengsui Decoction is an effective treatment for osteoporosis via PI3K-AKT.

Plant microRNA profiling of Honeysuckle and Honeysuckle Decoction

Project description:Influenza A viruses (IAVs), particularly H1N1, H5N1 and H7N9, pose a substantial threat to public health worldwide. Here, we report that MIR2911, a honeysuckle (HS)-encoded atypical microRNA, directly targets IAVs with a broad spectrum. MIR2911 is highly stable in HS decoction, and continuous drinking or gavage feeding of HS decoction leads to a significant elevation of the MIR2911 level in mouse peripheral blood and lung. Bioinformatics prediction and a luciferase reporter assay showed that MIR2911 could target various IAVs, including H1N1, H5N1 and H7N9. Synthetic MIR2911 significantly inhibited H1N1-encoded PB2 and NS1 protein expression but did not affect mutants in which the MIR2911-binding nucleotide sequences were altered. Synthetic MIR2911, extracted RNA from HS decoction and HS decoction all significantly inhibited H1N1 viral replication and improved viral infection-induced mouse weight loss but did not affect infection with a mutant virus in which the MIR2911-binding nucleotide sequences of PB2 and NS1 were altered. Importantly, the inhibitory effect of HS decoction on viral replication was abolished by an anti-MIR2911 antagomir, indicating that the physiological concentration of MIR2911 in HS decoction could directly and sufficiently suppress H1N1 viral replication. MIR2911 also inhibited H5N1 and H7N9 viral replication in vitro and in vivo. Strikingly, administration of MIR2911 or HS decoction both dramatically reduced mouse mortality caused by H5N1 infection. Our results demonstrate that MIR2911 is the first active component identified in Traditional Chinese Medicine to directly target various IAVs and may represent a novel type of natural product that effectively suppresses viral infection. The effects of traditional Chinese medicine Yinqi Sanhuang Jiedu Decoction on gene expression in HepG2.2.15 cells

Project description:Yinqi Sanhuang Jiedu Decoction (YQSHJD) is a clinically-proven formula for treating chronic hepatitis B and related fibrosis, demonstrating significant anti-HBV effects in clinical trials. However, its direct inhibitory effect on HBV and the underlying mechanisms remain unclear. The objective of this study is to elucidate the molecular mechanism through which YQSHJD inhibits HBV. HBV-replicating HepG2.2.15 cells were utilized as an in vitro HBV model to study the impact of YQSHJD on HBV replication. Network pharmacology and transcriptome analysis were used to research the targets for YQSHJD in inhibiting HBV replication. The results showed YQSHJD could inhibit HBV replication through regulation of the host antiviral targets.

Effect of Qingyi Decoction on the Treatment of Acute Pancreatitis Mice via mRNA Sequencing Analysis

Project description:Acute pancreatitis (AP) is an inflammatory disease of pancreatic tissue caused by pancreatic acinar cell injury. AP treatment of Qingyi Decoction(QYD) is a widely and commonly used traditional Chinese medicine prescription. A mice model of acute pancreatitis was established and treated with Qingyi Decoction, and mRNA high-throughput sequencing was performed on the pancreatic tissue of mice treated with Qingyi Decoction for the first time. There were 4477 and 4359 differentially expressed genes in the control group compared with the model group and the Qingyi Decoction group, while there were 8 differentially expressed genes in the model group compared with the Qingyi Decoction group by analyzing the mRNA expression profiles. Analysising of KEGG, GO, GSEA and network interaction, it was found that its therapeutic effect was closely related to apoptosis, inflammatory response and autophagy process.

Integrative Pharmacology Powers the Active Components Detection and Mechanism Exploration of Wang Bi Granules Acting on Rheumatoid Arthritis

Project description:Wang Bi Granule (WBG), composed of 16 botanical drugs and 1 animal drug, is clinically used to treat “Wang Bi” syndrome, which mainly refers to later rheumatoid arthritis (RA) in modern medicine, as well as ankylosing spondylitis, tuberculous arthritis, and Kashin-Beck disease characterized by joint pain and swelling deformation. However, its chemical composition and pharmacological mechanisms have not been clarified. Therefore, in the present study we aimed to characterize the chemical components of WBG and explore its mechanism in treating RA using integrative pharmacology, including chemical composition detection, efficacy evaluation, and mechanism exploration. We employed UPLC-QTOF-MS/MS to describe the chemical profile of WBG, TNF-α-stimulated RAW264.7 cells to simulate inflammatory process of RA and evaluate the anti-inflammatory effect of WBG, and network pharmacology to mine the mechanism of WBG acting on RA. As a result, a total of 278 chemical components were identified or tentatively characterized and the water extract of WBG improved the imbalance of inflammation by regulating 179 differential genes in RAW264.7 cells induced by TNF-α. 55 key active constituents were obtained based on the interactions among “components’ targets, RA-related genes, and differential genes (WBG vs TNF-α group)”, which may ameliorate RA conditions by regulating 161 hub genes that mainly involved in inflammation-immune related pathways. The present study, for the first time, employed integrative pharmacology to characterize the chemical profile of WBG and reveal its mechanism against RA through inflammation-immune regulation system.

Chinese Herbal Formula (GCNY)-Medicated Serum Alleviates Peroxidation Induced by H2O2 in Human Microglial Cells

Project description:Transitional Chinese herbal medicine has a long history of applications for anti-aging and longevity purposes, and in our patent formula Gancao Nurish-Yin (GCNY) decoction, all of the ingredients show antioxidant properties. However, in real clinical practice, extractions of herbs are rarely applied alone but are prescribed as integrated formula. In order to investigate whether GCNY possess anti-oxidation potential, we applied GCNY as a formula to treat rats in order to acquire medicated serum, which was then added on H2O2 (200 μM)-modeled human microglial cell line HMC-3 in comparison with its controls.

Proteomics reveals the mechanism of Qingre Huo xue decoction in treating rheumatoid arthritis

Project description:Rheumatoid arthritis is an autoimmune disease characterized by chronic synovitis, which can lead to irreversible joint damage as well as cardiovascular and respiratory system damage. The Qingre Huoxue Decoction is a Chinese medicine compound that has been clinically proven to be effective in treating RA, but its mechanism has not yet been elucidated. This study identified 83 differentially expressed proteins, including 46 upregulated proteins and 37 downregulated proteins, through proteomic analysis of the serum of RA patients before and after 24 weeks of treatment with Qingre Huoxue Decoction and healthy human. Among them, FBP1 protein may be a potential therapeutic target of Qingre Huoxue Decoction.
