Single cell RNA-seq analysis revealed molecular regulation of hematopoietic stem cell fate decisions in response to hypoxia

ABSTRACT: Here, we conducted single-cell RNA Seq to establish the atlas of hematopoietic stem progenitor cells (HSPC) and acquire the difference between the choice of cell fate.This study reports how the hypoxic condition influences the HSPCs, revealing the fate of erythroid and monocyte lineage. The immunomodulatory of MSCs was also affected, enhancing the development of monocytes. In addition, stem cells maintain quiescence in hypoxia. This would contribute to understanding regulatory mechanisms for hematopoiesis under a hypoxic state, favouring our comprehension of perturbation, facilitating the manipulation of cell-based therapies.
