Project description:Prostate cancer DU145 tumors were subcutaneously established in male nude mice. In nude mice, Zelnorm were orally administrated. Tumor volume and body weight were assessed (tumor volume=1/2 (length×width2)). Tumors were excised and weighted. Tumor samples were profiled by RNA-seq analysis. Respectively, to differentiate the gene expression originated from human (cancer cells) or mouse (tumor TME part), data were mapped against human (GRCh38) and mouse (GRCm38) genomes separately.

Project description:We performed survival analysis of control and MCD groups, and explored underlying tumor suppression mechanisms after dietary intervention, focused on alterations in the energy-dependent signaling pathways, histone modifications, and global gene expression differences on cDNA microarray study. Illumina arrays: Five- week- old male C57BL6 mice were randomly divided into two groups and fed control diet (control group, LabDiet, Brentwood, MO, USA) or moderate restriced carbohydrate diet formula (MCD, Treat group) in a specific pathogen free zone. All procedures were approved by the institutional animal use and care committee. Following a preliminary feeding of each diet formula for one week, 1 × 106 B16F10 cells (suspended with 100 μl of PBS) were subcutaneously injected into the back of the mice. After 2 weeks of diet supplementation, all mice were sacrificed. Tumor tissue was excised for cDNA microarray. Project description:Four vehicle-treated and four HhAntag-treated pancreatic xenograft tumors were profiled for gene expression changes using Affymetrix U133 Plus 2.0 and Affymetrix Mouse Genome 430 2.0 arrays. Keywords: comparative gene expression, hedgehog, hh A primary human pancreatic tumor xenograft (1051178-A) was established by direct implantation of surgical material into female CD1 nu/nu mice of 6-8 weeks of age. Tumors were serially passaged into larger cohorts of mice for efficacy testing and subsequently distributed into tumor volume-matched cohorts upon tumors reaching between 200 to 350 mm3. HhAntag was resuspended in 0.5% methyl-cellulose, 0.2% Tween-80 (MCT) and administered orally twice daily at 75 mg/kg from a 10 mg/ml suspension. MCT alone served as vehicle control. Tumor xenografts (4/group) were excised following 21 days of dosing and RNA was extracted. Preparation of complementary RNA, Human Genome U133 Plus 2.0 array and Mouse Genome 430 2.0 array hybridizations, and subsequent data analysis were carried out using Affymetrix protocols, with signal intensities being determined by the MAS5.0 algorithm.

Project description:RNA from tumors treated with vehicle or 30 mg/kg GTx-027 were pooled and subjected to microarray analysis. Genes that were increased or decreased by 2-fold or more were considered for further analyses. Unlike in prostate cancer, where AR agonists induce more genes than they repress, in MDA-MB-231-AR tumors, GTx-027 inhibited 2.5X the number of genes (1092 vs. 456) than it activated. Functional clustering of the genes indicated that GTx-027 modified more breast cancer genes than other pathway genes. Genes that regulate the function of others cancers, such as colorectal, lung, and oral, and metabolic diseases were also favorably altered by GTx-027. Breast cancer proliferative genes, such as aurora kinase, ERCC1, IGFBP3 were inhibited and growth inhibitory genes, such as NQO1, PTPRJ were activated by GTx-027. Many of the established androgen responsive-genes were also activated by GTx-027, indicating that breast cancer growth inhibitory role of GTx-027 evolved from its agonistic activity. A mixture of cells was suspended in 0.05 ml RPMI+10% FBS and 0.05 ml Matrigel/animal and was injected subcutaneously. Once the tumor size reached 200-300 mm3, the animals were randomized and treated orally with the indicated drugs formulated in Tween 80:Captex 200:water (0.8:0.2:9). Tumor volume and body weight were measured thrice weekly. At the end of study, animals were sacrificed, tumors excised, weighed, and stored for various analyses. RNA from tumors was isolated and verified qualitatively and quantitatively. Samples from each group (n=8; 100 ng/µl; total 1000 ng) were pooled and hybridized to Affymetrix human ST2.0 gene array. Data from the array were analyzed using Ingenuity Pathway Analysis software (IPA3).

Project description:Global gene expression profiling reveals a potential anti-melanoma effect of SQ-diEG in B16F10 lung colonization mice model. Squalene (SQ) was considered as a promising natural agent in anti-cancer treatment due to its strong antioxidant and anti-inflammatory activity; however, its pharmacological value has been largely underestimated because of its poor solubility and bioavailability. To adress this problem, a novel amphiphilic SQ derivative which bearing ethylene glycol oligomers was synthesized and was used as a permeation enhancer in this study to check its potential effect on anti-melanoma metastasis using B16F10 lung colonization mice model. Total 40 six years old male C57BL/6J mice were randomly divided into five groups (8 mice per group), including the control "(-)B16F10 injected and water treated"; the model "(+) B16F10 injected and water treated"; the positive control "(+) B16F10 injected and DTIC treated (Dacarbazine, 70 mg/kg/day)"; the SQ-diEG treatment "(+) B16F10 injected and SQ-diEG treated (5 mg/kg/day)"; the SQ-diEG treatment "(+) B16F10 injected and SQ-diEG treated (25 mg/kg/day)". After one week of acclimatization, B16F10 were murine melanoma cells line which intravenously injected (200000 tumor cells in 100 μl PBS, 0.2 ml/mouse) through the tail vein of mice. Mice was orally administrated with Dacarbazine (DTIC, positive control, 70mg/kg), and SQ-diEG (5mg/kg and 25mg/kg) for 20 days. On the last day of administration, mice was sacrificed and its lung was washed and collect for the further RNA extraction. Finally, the mice microarray gene expression profiling was conducted for two biological replicates.

Project description:The effects of AKT/WEE1 inhibitors or ALDH inhibitor combined with PD-1 antibody on melanoma were examined. Mouse B16F10 melanoma cells were subcutaneously injected into C57/B6 mice. When tumors were well-vascularized, mice were treated with anti-PD-1, AKT I (AZD5363)+WEE1 I (MK1775), AZD5363+MK1775+anti-PD-1, ALDH I (KS100), KS100+ anti-PD-1 daily. Mice were sacrificed when tumors in the control group (DMSO treated) reached 2000 mm3 and tumors were removed. RNA was extracted from tumor samples and gene expression was analized using RNAseq analysis.
