Identification of exercise-related genes in mice exposed to cigarette smoke

Project description:Cigarette smoke (CS) is the major risk factor for COPD and is linked to cardiopulmonary dysfunction. Exercise training, as part of pulmonary rehabilitation, is recommended for all COPD patients. It has several physiological benefits, but the involved mechanisms remain poorly defined. Here, we employed transcriptomic profiling and examined lung endothelium to investigate novel interactions between exercise and CS on cardiopulmonary alterations. Mice were exposed to 20 weeks of CS, CS + 6 weeks of high-intensity interval training on a treadmill or control. Lung and cardiac (left and right ventricle) tissue were harvested, and RNA-sequencing was performed and validated with RT-qPCR. Immunohistochemistry assessed pulmonary arteriolar changes. Transcriptome analysis between groups revealed 37 significantly regulated genes in the lung, 21 genes in the left ventricle, and 43 genes in the right ventricle (likelihood-ratio test). Validated genes that showed an interaction between exercise and CS included angiotensinogen (p=0.002) and resistin-like alpha (p=0.019) in left ventricle, with prostacyclin synthetase different in pulmonary arterioles (p=0.004). Transcriptomic profiling revealed changes in pulmonary and cardiac tissue following exposure to CS, with exercise training exerting rescue effects. Exercise-regulated genes included angiotensinogen and resistin-like alpha. However, it remains unclear if these represent potential candidate genes or biomarkers involved during pulmonary rehabilitation.

Effects of Voluntary Exercise on Hippocampal Gene Expression in male C57Bl/6J mice

Project description:This experiment was designed to identify changes in gene expression in the brain (specifically the hipocampus) in relation to variable levels of voluntary exercise. Male C57Bl/6J mice were weaned onto a low fat diet (LF; Research Diets D12329, 11% kcal fat) and given continuous access to exercise wheels (30.3 x 20.6 x 26 cm; Mini Mitter®, Nalgene® Activity Wheel for Rodents) at ten weeks of age, for three days (n=96). Six mice had their wheels locked to act as sedentary controls (sed). Animals were considered to be high runners (HR) if they ran >= 5 km/night and low runners (LR) if they ran <= 2 km/night. Middle runners (MR) were chosen randomly from the population of animals falling between the high and low groups. This series is comprised of array results from three parallel experiments comparing HR versus sed, MR versus sed, and HR versus LR. Six hybridizations were performed for each of the three comparisons (3 forward-labeled, 3-reverse labeled) to account for dye-incorporation differences. One slide from the HR vs LR experiment was broken during processing, so only five are included from that group. Transcription profiling of rat heart left ventricle after exercise to investigate the role of exercise as a known protective factor against cardiovascular morbidity and mortality

Project description:Physical exercise training is a known protective factor against cardiovascular morbidity and mortality. Nevertheless, the underlying specific molecular mechanisms still remain uncompletely explored. To identify molecular mechanisms by which exercise training induces this favorable phenotype a genomic approach was used in an animal model of mild exercise previously demonstrated by our group to induce cardioprotection. Our data indicate that mild exercise by inducing no or few permanent changes in gene expression profile is able to determine cardioprotection without induction of cardiac hypertrophy. Experiment Overall Design: We investigated the gene expression profile by Affymetrix technology (230 2.0 GeneChip rat genome array) induced by mild exercise training (treadmill running: 25 m/min, 10% incline, 1 h/day, 3 days/week, 10 weeks) on left ventricle (LV) of exercise-trained (n=10) and sedentary control (n=10) rats.

eNOS interactors in human cells

Project description:Endothelial nitric oxide synthase (eNOS) catalyzes the conversion of L-arginine and molecular oxygen into L-citrulline and nitric oxide (NO), a gaseous second messenger that influences cardiovascular physiology and disease. Several mechanisms regulate eNOS activity and function, including phosphorylation at Ser and Thr residues and protein-protein interactions. Combining a tandem affinity purification approach and mass spectrometry, we identified stromal cell-derived factor 2 (SDF2) as a component of the eNOS macromolecular complex in endothelial cells. SDF2 knockdown impaired agonist stimulated NO synthesis and decreased phosphorylation of eNOS at Ser1177, a key event required for maximal activation of eNOS. Conversely, SDF2 overexpression dose-dependently increased NO synthesis through a mechanism involving Akt and calcium (induced with ionomycin), which increased the phosphorylation of Ser1177 in eNOS. NO synthesis by iNOS (inducible NOS) and nNOS (neuronal NOS) was also enhanced upon SDF2 overexpression. We found that SDF2 was a client protein of the chaperone protein Hsp90, interacting preferentially with the M domain of Hsp90, which is the same domain that binds to eNOS. In endothelial cells exposed to vascular endothelial growth factor (VEGF), SDF2 was required for the binding of Hsp90 and calmodulin to eNOS, resulting in eNOS phosphorylation and activation. Thus, our data describe a function for SDF2 as a component of the Hsp90-eNOS complex that is critical for signal transduction in endothelial cells.

Gene expression for eNOS S1176 mutant mice

Project description:Extensive reports in the past decade have shown the importance of eNOS expression in atherogenesis. However, both the eNOS knockout and overexpression mouse models exhibit paradoxical results of increased atherosclerotic lesions. Our genetic approach herein utilizes endogenous point-mutations at the eNOS S1176 site to effectively manipulate eNOS activity rather than total eNOS protein expression levels. Our efforts definitively show that eNOS S1176 phosphorylation is sufficient to mitigate atherosclerotic lesion progression.

Age-dependent impact of two exercise training regimens on genomic and metabolic remodeling in skeletal muscle and liver of male mice

Project description:How skeletal muscle adapts to different types of exercise intensity with age is not known. Adult and old C57BL/6 male mice were assigned to one of three groups: sedentary, daily high-intensity intermittent training (HIIT), or moderate intensity continuous training (MICT) for 4 weeks, compatible with the older group’s exercise capacity. Improvements in body composition, fasting blood glucose, and muscle strength were mostly observed in the MICT old group, while effects of HIIT training in adult and old animals was less clear. Skeletal muscle exhibited structural and functional adaptations to exercise training, as revealed by electron microscopy, OXPHOS assays, respirometry, and muscle protein biomarkers. Transcriptomics analysis of gastrocnemius muscle combined with liver and serum metabolomics unveiled an age-dependent metabolic remodeling in response to exercise training. These results support a tailored exercise prescription approach aimed at improving health and ameliorating age-associated loss of muscle strength and function in the elderly.
