RNA-seq analysis of genes differentially expressed in NCI-H1299 cells with platensimycin (PTM) derivative 6p treatment

ABSTRACT: In order to characterize the global transcriptional effect and evaluate the impact of PTM derivative 6p on non-small cell lung cancer NCI-H1299 cell gene expression. NCI-H1299 cells were treated with vehicle (0.1% DMSO) or 25 μM 6p for 48 h and assessed the gene expression pattern by RNA-Seq. Finally, we show that extensive, coordinated effects of 6p on the transcriptional activity of genes within pathways controlling tumor cell metabolism, proliferation, and survival. Especially, the expression of key genes in de novo lipid synthesis were significantly reduced.
