MRNA sequencing of primary tumors resected from mice

ABSTRACT: Mice were injected with MDAMB231 cells (weakly adherent, strongly adherent, or unsorted control) that were allowed to grow into a tumor for 6 weeks. After 6 weeks, the mice were sacrificed and the tumors were resected. mRNA sequencing was performed on dissociated cells from the tumors.

Ager (Rage) and Myd88 deletion in resected mouse livers

Project description:The effect of resection of mouse livers on survival serve as a model for the effect of resection of human livers, which are performed for both the purposes of removing sick tissue and for transplanting healthy tissue. When less than 70% of a mouse liver is resected, the mouse usually lives. However, when more than 70% is resected, there is an increased probability that the mouse will die. We have found that deletion of Ager (Advanced Glycation End products receptor) (Rage) increases the chance of mouse survival relavtive to WT. Conversely, the deletion of Myd88 decreases the chances of survival, as does the simultaneous deletion of both Ager and Myd88. To understand these genotype/phenotype relations, we performed gene expression measurements. Gene expression of livers of WT C57BL/6 mice, Ager-null C57BL/6 mice, Myd88-null C57BL/6 mice, and Ager-null/Myd88-null C57BL/6 mice, all 10-12 weeks-old and sacrificed 2 hours after 85% liver resection, were compared. 4-6 replicates each group.

Tumor Heterogeneity in Metastatic Potential is Indicated by Adhesion Strength

Project description:Tumors are heterogeneous and comprised of cells with varying function and ability to disseminate. Despite significant effort, no universal biological marker currently serves as a metric for metastatic potential of solid tumors. Common to disseminating cells from such tumors, however, is the need to modulate their adhesion as they detach from the tumor and migrate through stroma to intravasate. Adhesion strength is heterogeneous even amongst cancer cells within a given population, and using a parallel plate flow chamber, we separated and sorted these populations into weakly and strongly adherent groups; when cultured under stromal conditions, this adhesion phenotype is stable over multiple days, sorting cycles, and common across all epithelial tumor lines investigated. Subpopulations do not show differences in expression of proteins involved in the focal adhesion complex but do exhibit intrinsic focal adhesion assembly as well as contractile differences that result from differential expression of genes involved in microtubules, cytoskeleton linkages, and motor activity. Collectively, these differences drive migration in both 2D and 3D migration assays for weakly adherent cells; these differences are maintained over days in culture, suggesting that adhesion strength can serve as a stable marker for migration and metastatic potential within a given tumor population. Moreover, these data suggest that the fraction of weakly adherent cells present within a tumor could act as a novel physical marker for metastatic potential.

Expression data from the orthotopic tumors of MES-SA

Project description:The spontaneous pulmonary metastasis model of human uterine sarcoma was established using GFP-expressed MES-SA cells. Several sublines with different metastatic potentials were generated by in vivo passaging. We used microarrays to identify the metastatic-related genes using the orthotopic tumors with different metastatic potentials. Orthotopic transplanted uterine sarcoma were observed after 3 weeks post-transplantation and the xenografted tumor reached about 2000 mm3 in additional 3 weeks. After 6 weeks post-transplantation, the animals were sacrificed under isoflurane anesthesia, and the tumors were resected. The GFP-positive regions were collected using Leica MZ10F fluorescent stereomicroscope. This study compared the gene expression profiling between the orthotopic tumors with high (H)- and low (L)-metastatic potentials.

Once serum AFP levels reached levels consistent with tumor growth of ~80-100 mg, animals were subjected to a single IV injection of MRX34. Twenty-four hours later animals were sacrificed, tumors resected and RNA isolated for subsequent microarray analysis.

Project description:We devised a novel methodology for sorting stroma-free tumor cells according to their relative distance from BVs by injecting a vascular perfusion marker (Hoechst 33342). Briefly, post 3 weeks of intracranial implantation of U87MG-GFP cell line into striatum of NOD-SCID mice, the mice were injected i.v. with Hoechst dye. Mice sacrificed, tumors retrieved, single cell dissociated and FACS sorted to isolate cells that are either Hoechst Positive (Hh) or Hoechst Negative (Hl). RNA was extracted from the sorted cells and RNA-Sequencing performed.
