ABSTRACT: MDAMB231 cells express the SV40TAg signature. The normal mammary epithelial tissue does not express the signature. CK0082, Empty1 and Gata3 samples are not reported in this manuscript We used microarrays to determine whether the MDAMB231 cell lines and tumors express the SV40Tag signature (91 gene signature).

Project description:M6 cells expression a similar genetic signature as the parent tumor, C3(1)Tag tumor. The mammary tumors, and tumor cell line, are distinct from normal mammary epithelial tissue (FVB) BRCA/p53, cmyc, h2n, p53ERneg, p53ERpos, pymt and ras samples were not reported in this paper. We used microarrays to determine whether the M6 cell line retains the SV40Tag signature (91 gene signature) that is expressed in C3(1)Tag tumors but not FVB normal mammary tissue.

Project description:After 7 days of culture, a sub-population of breast cancer MDAMB231 cells spontaneously detaches the monolayer and starts to grow in suspension, these cells undergo epithelial-mesenchymal transition (EMT), display anoikis resistance and acquire a metastatic phenotype. These cells, in the presence of adherently growing MDAMB231, continue to grow in suspension whereas, seeded in cell-free wells, are able to adhere again and to form E-cadherin positive and vimentin negative new colonies, suggesting the occurrence of mesenchymal-epithelial transition (MET). The expression profiles of these three cell clones (basal, MET and EMT) were investigated by microarray analysis.

Comparison between hereditary breast tumors and normal breast tissue samples.

Project description:Tumor associated miRNAs in hereditary breast cancer. In this study we investigated the role of miRNAs in hereditary breast tumors comparing with normal breast tissue. Global miRNA expression profiling was performed on 22 hereditary breast tumors and 15 non-tumoral breast tissues. We identified 19 miRNAs differentially expressed, most of them down-regulated in tumors. An important proportion of deregulated miRNAs in hereditary tumors were previously identified commonly deregulated in sporadic breast tumors. Our results identify miRNAs associated to hereditary breast cancer, as well as miRNAs commonly miss-expressed in hereditary and sporadic tumors, suggesting common underlying mechanisms of tumor progression. In addition, we provide evidence that KRAS is a target of miR-30c, and that this miRNA suppresses breast cancer cell growth potentially through inhibition of KRAS signaling. Selectivity profiling of SLC16A3/MCT4 inhibitor slCeMM1 using photoaffinity probe

Project description:To assess the selectivity of SLC16A3/MCT4 inhibitor slCeMM1, we derived diazirine-alkyne photoaffinity probe (slCeMM1-PAP). We next used HAP1 and MDAMB231 cell lines for MS experiment, comparing the enrichment of proteins by slCeMM1-PAP and conditions where slCeMM1-PAP was competed with slCeMM1 or it’s structurally related inactive control (S1_007). We found that among proteins enriched by slCeMM1-PAP, only SLC16A3 was competed by slCeMM1, but not S1_007, confirming the SLC16A3 engagement by slCeMM1 and suggesting overall good selectivity of slCeMM1.

Transcription profiling of human mesenchymal stem cells reveals carcinoma associated fibroblast like differentiation

Project description:Carcinoma associated fibroblasts (CAFs) have recently been implicated in important aspects of epithelial solid tumor biology such as neoplastic progression, tumor growth, angiogenesis, and metastasis. However, neither the source of CAFs nor the differences between CAFs and fibroblasts from non-neoplastic tissue have been well defined. In this study we demonstrate that human bone marrow-derived mesenchymal stem cells (hMSCs) exposed to tumor-conditioned medium (TCM) over a prolonged period of time assume a CAF-like myofibroblastic phenotype. More importantly, these cells exhibit functional properties of CAFs including sustained expression of stromal derived factor 1 (SDF-1) and the ability to promote tumor cell growth both in vitro and in an in vivo co-implantation model and expression of myofibroblast markers including alpha-smooth muscle actin and fibroblast surface protein. hMSCs induced to differentiate to a myofibroblast-like phenotype using 5-azacytidine do not promote tumor cells growth as efficiently as hMSCs cultured in tumor-conditioned medium nor do they demonstrate increased SDF-1 expression. Furthermore, gene expression profiling revealed similarities between TCM exposed hMSCs and carcinoma associated fibroblasts. Taken together these data suggest that hMSCs are a source of carcinoma associated fibroblasts and can be used in the modeling of tumor-stroma interactions. To our knowledge this is the first report demonstrating that hMSCs become activated and resemble carcinoma associated myofibroblasts upon prolonged exposure to conditioned medium from MDAMB231 human breast cancer cells. Experiment Overall Design: The experiment was done to test the differentiation of hMSCs upon exposure to tumor condition media or epigenetic modifiers.

Gene expression analysis of the immortalized human endothelial cell lines HMEC-1 and TIME

Project description:The distinction between lymphatic and blood vessels is biologically fundamental. Two immortalized cell lines, which have been widely used as models for endothelial cells of blood vascular origin, are the human microvascular endothelial cell line-1 (HMEC-1) and the telomerase-immortalized microvascular endothelial cell line (TIME). However, analysis of protein expression by flow cytometry revealed expression of lymphatic markers on these cell lines. Furthermore, functional in vitro leukocyte transmigration assays demonstrated deficiencies in several steps of the leukocyte extravasation cascade. Hence we performed this microarray analysis of the gene expression in HMEC-1 and TIME. We then compare the expression profiles to those of published blood- and lymphatic endothelial cells. Analysis of the gene expression profile of the immortalized human endothelial cell lines HMEC-1 and TIME to be compared to published primary blood- and lymphatic endothelial cells (see supplementary file linked below).

Time course for bladder UCC development in UPII-SV40Tag mice

Project description:We have identified genes that are differentially expressed between the bladders of UPII-SV40Tag mice and their age-matched wild-type littermates at 3, 6, 20, and 30 weeks of age. These are ages that correspond to premalignant, carcinoma in situ, and early-stage and later stage invasive UCC, respectively Analysis of the microarray data sets has revealed ∼1,900 unique genes differentially expressed (≥3-fold difference at one or more time points) between wild-type and UPII-SV40Tag urothelium during the time course of tumor development.
