Nuclear actin assembly is needed for decidualization of human endometrial stromal cells

3’,5’-cAMP affects actin cytoskeleton dynamics by direct binding to actin.

Project description:Cyclic adenosine monophosphate (3’,5’-cAMP) is a well-characterized and evolutionary conserved second messenger. In contrast to the animal cells, however, the role of 3’,5’-cAMP in plants remains enigmatic. With the exception of cyclic nucleotide-gated ion channels, no plant 3’,5’-cAMP interactors have been reported to date. To address the existing gap, we resorted to thermal proteome profiling (TPP) as novel biochemical approach for the identification of protein binders of a small-molecule of choice. Based on the significant change in the thermal stability caused by 3’,5’-cAMP, we report a list of 51 putative 3’,5’-cAMP protein interactors, of which we focused on actin isovatriant ACT2. Most importantly we could demonstrate that 3’,5’-cAMP binding increases the rate of actin polymerization, resulting in longer actin filaments. Moreover, 3’,5’-cAMP treatment affected actin bundling, resulting in thicker actin bundles, as observed in the Arabidopsis hypocotyl cells. In line, with the obtained results 3’,5’-cAMP supplementation resulted in partial rescue of the short hypocotyl phenotype of the act2act7 Arabidopsis mutant, and 3’,5’-cAMP treatment suppressed actin depolymerization induced by Latrunculin B. Finally, 3’,5’-cAMP effects were independent from its degradation to adenosine, and true for nanomolar concentrations of 3’,5’-cAMP reported for the plant cells. Considering evolutionary conservation of 3’,5’-cAMP and actin we expect that our findings will be relevant to other eukaryotic organisms, and as such demonstrate a novel and direct mechanism of 3’,5’-cAMP regulation of the actin cytoskeleton dynamics.

TFAM loss induces nuclear actin assembly upon mDia2 malonylation to promote liver cancer metastasis

Project description:The mechanisms underlying cancer metastasis remain poorly understood. Here, we report that TFAM deficiency rapidly and stably induced spontaneous lung metastasis in mice with liver cancer. Interestingly, unexpected polymerization of nuclear actin was observed in TFAM-knockdown HCC cells when cytoskeleton was examined. Polymerization of nuclear actin is causally linked to the high-metastatic ability of HCC cells by modulating chromatin accessibility and coordinating the expression of genes associated with extracellular matrix remodeling, angiogenesis, and cell migration. Mechanistically, TFAM deficiency blocked the TCA cycle and increased the intracellular malonyl-CoA levels. Malonylation of mDia2, which drives actin assembly, promotes its nuclear translocation. Importantly, inhibition of malonyl-CoA production or nuclear actin polymerization significantly impeded the spread of HCC cells in mice. Moreover, TFAM was significantly downregulated in metastatic HCC tissues and was associated with overall survival and time to tumor recurrence of HCC patients. Taken together, our study connects mitochondria to the metastasis of human cancer via uncovered mitochondria-to-nucleus retrograde signaling, indicating that TFAM may serve as an effective target to block HCC metastasis.

Project description:Function and fate of mRNAs is controlled by RNA binding proteins (RBPs) but determining the proteome of a specific mRNA in vivo is still challenging. RNA proximity biotinylation on the transported β-actin mRNA tagged with MS2 aptamers (RNA-BioID) is used to characterize the dynamic proteome of the β-actin mRNP in mouse embryonic fibroblasts (MEFs). We have identified > 60 β-actin associated RBPs including all six previously known as well as novel interactors. By investigating the dynamics of the β-actin mRNP in MEFs, we expand the set of β-actin mRNA associated RBPs and characterize the changes of the interacting proteome upon serum-induced mRNA localization. We report that the KH-domain containing protein FUBP3 represents a new β-actin associated RBP that binds to its 3’-untranslated region outside the known RNA localization element but is required for β-actin RNA localization. RNA-BioID will allow to obtain a dynamic view on the composition of endogenous mRNPs.
