Single cell RNA-seq in chicken's lung and DF-1 cell line

ABSTRACT: Newcastle disease virus (NDV) is an avian paramyxovirus that causes major economic losses to the poultry industry around the world, with NDV pathogenicity varying due to strain virulence differences. However, the impact of intracellular viral replication and the heterogeneity of host responses among cell types are unknown. Here, we investigated the heterogeneity of lung tissue cells in response to NDV infection in vivo and that of chicken embryo fibroblast cell line DF-1 to NDV infection in vitro using single-cell RNA sequencing.

RNA-Seq Analysis of Spleen from Chicken and Japanese Quail infected with Lentogenic and Velogenic Newcastle Disease Virus

Project description:Newcastle disease (ND) affects a few hundred avian species including chicken, and the clinical outcome of Newcastle disease virus (NDV) infection ranges from mild to severe fatal disease depending on the NDV pathotype and the host species involved. Japanese quails serve as natural reservoirs of NDV and play important role in NDV epidemiology. While infection of chicken with velogenic NDV results in severe often fatal illness, the same infection in Japanese quails is results in in apparent infection. The molecular basis of this contrasting clinical outcomes of NDV infection is not yet known. We compared global gene expression in spleens of chicken and Japanese quails infected with a lentogenic or velogenic NDVs. We found contrasting regulation of key genes associated with NF-κB pathway and T-cell activation between chicken and Japanese quails. Our data suggests association of NDV resistance in Japanese quails to activation of NF-κB pathway and T cell proliferation.

In Vitro and In Vivo Metabolomic Profiling after Infection with Virulent Newcastle Disease Virus

Project description:Newcastle disease (ND) is an acute, febrile, and highly contagious disease caused by the virulent Newcastle disease virus (vNDV) worldwide. The disease causes serious economic losses to the poultry industry. However, the metabolic changes caused by vNDV infection remain unclear. The objective of this study was to determine whether small molecule metabolites contribute to the pathogenesis of vNDV. DF-1 cells and the lungs of specific pathogen free (SPF) chickens infected with the vNDV strain Herts/33 were analyzed via ultra high-performance liquid tandem chromatography quadrupole time of flight mass spectrometry (UHPLC-QTOF-MS) in combination with multivariate statistical analysis. A total of 304 metabolites were significantly changed post vNDV infection, and most belong to the amino acid and nucleotide metabolic pathway. It is suggested that the increased pools of amino acids and nucleotides may be used for viral protein synthesis and genome amplification. Similar results were also confirmed in vivo; this is consistent with the characteristic of acute vNDV infection. The identification of these different metabolites will provide a great deal of important information to further understand the mechanism of vNDV replication and pathogenesis.

Oligoadenylate synthetase-like (OASL) aggravated newcastle disease virus (NDV)-induced necroptosis in glioma cells

Project description:Newcastle disease virus (NDV) has emerged as an oncolytic agent in several cancers. Previous study has shown that NDV exerts cytolytic activity in glioma, however, the underlying mechanism has not been fully uncovered. Here the cytolytic activity of NDV in glioma and the associated mechanisms have been demonstrated. Infection with NDV inhibits cell proliferation and promotes cell apoptosis in LN229 cells. Further investigation showed that cytoplasmic organelle damage and cytoplasmic vacuolation were observed in LN229 cells after NDV infection. JC-1 staining assay proved that NDV caused cell apoptosis of LN229 cells by inducing mitochondrial dysfunction. We next speculated that NDV caused LN229 cells death through inducing necroptosis, but not ferroptosis, since the Fe2+ level did not alter after NDV infection. Furthermore, the NDV-caused cell death in LN229 cells was blocked by necroptosis inhibitor Nec1. Besides, RNA-seq analysis identified the different expression genes in NDV-infected LN229 cells. OASL, an antiviral gene, has been found to be directly induced by NDV infection. We also found that knockdown of OASL enhanced NDV infection-induced LN229 cells necroptosis. In summary, two aspects about cytolytic activity of NDV in glioma have been demonstrated. NDV presented cytolytic activity in glioma cells through inducing necroptosis. Additionally, targeting OASL may provide new strategy for enhancing necroptosis of glioma cells after NDV infection.

Project description:Hanl Lee & Angelyn Lao. Transmission dynamics and control strategies assessment of avian influenza A (H5N6) in the Philippines. Infectious Disease Modelling 3 (2018). Due to the outbreaks of Highly Pathogenic Avian Influenza A (HPAI) H5N6 in the Philippines (particularly in Pampanga and Nueva Ecija) in August 2017, there has been an increase in the need to cull the domestic birds to control the spread of the infection. However, this control method poses a negative impact on the poultry industry. In addition, the pathogenicity and transmissibility of the H5N6 in both the birds and the humans remain largely unknown which call for the necessity to develop more strategic control methods for the virus. In this study, we constructed a mathematical model for the bilinear and half-saturated incidence to compare their corresponding effect on transmission dynamics of H5N6. The simulations of half-saturated incidence model were similar to what occurred during the H5N6 outbreak (2017) in the Philippines. Instead of culling the birds, we implemented other control strategies such as non-medicinal (personal protection and poultry isolation) and medicinal (poultry vaccination) ways to prevent, reduce, and control the rate of the H5N6 virus transmission. Among the proposed control strategies, we have shown that the poultry isolation strategy is still the most effective in reducing the infected birds.
