Project description:BackgroundAbdominal obesity is appreciated as a major player in insulin resistance and metabolically dysfunctional adipose tissue. Inappropriate extracellular matrix (ECM) remodelling and functional alterations in human adipose stromal/stem cells (hASCs) have been linked with visceral white adipose tissue (vWAT) dysfunction in obesity. Understanding the interactions between hASCs and the native ECM environment in obese vWAT is required for the development of future therapeutic approaches for obesity-associated metabolic complications.MethodsThe phenotypes and transcriptome properties of hASCs from the vWAT of obese patients and lean donors were assessed. The hASC-derived matrix from vWAT of obese or lean patients was generated in vitro using a decellularized method. The topography and the major components of the hASC-derived matrix were determined. The effects of the obese hASC-derived matrix on cell senescence and mitochondrial function were further determined.ResultsWe showed that hASCs derived from the vWAT of obese patients exhibited senescence and were accompanied by the increased production of ECM. The matrix secreted by obese hASCs formed a fibrillar suprastructure with an abundance of fibronectin, type I collagen, and transforming growth factor beta 1 (TGF-β1), which resembles the native matrix microenvironment of hASCs in vWAT derived from obese patients. Furthermore, the obese hASC-derived matrix promoted lean hASC ageing and induced mitochondrial dysfunction compared to the lean hASC-derived matrix. Blockade of TGF-β1 signalling using an anti-TGF-β1 neutralizing antibody alleviated the lean hASC senescence and mitochondrial dysfunction induced by the obese hASC-derived matrix.ConclusionsNative ECM in obesity vWAT initiates hASC senescence through TGF-β1-mediated mitochondrial dysfunction. These data provide a key mechanism for understanding the importance of cell-ECM interactions in hASCs senescence in obesity.

Human visceral adipose tissue RT-PCR in individuals with obesity

Project description:Visceral adiposity is more related with insulin resistance than BMI itself. To study the molecular signature of the subsequent evolution steps of the metabolic sequalae of obesity we collected omental adipose tissue samples from individuals with obesity while undergoing bariatric surgery. Participants (all with obesity) were divided in 4 groups according to glycemic profile: Insulin sensitive/normoglycemic group (IS NG) - OxHOMA2<1 and fasting glycemia < 100 mg/dL (n=17); Insulin resistant/normoglycemic group (IR NG) - OxHOMA2 > 1 and fasting glycemia < 100 mg/dL and HbA1c < 5.7% (n=24); Pre diabetes group (PD) - OxHOMA2 > 1 and fasting glycemia > 100 mg/dL or HbA1c [5.7% - 6.4%] (n=33); Type 2 diabetes group (T2D) - OxHOMA2 > 1 and fasting glycemia > 125 mg/dL or HbA1c > 6.4% (n=18). A panel of 43 genes related to inflammation, angiogenesis, lipid metabolism and redox machinery was evaluated in a BioMark HD Fluidigm system.

TGFβ1 determines the fate of human adipose-derived stromal cells by restraining their immaturity and white/beige adipocyte potentials

Project description:Human adipose-derived stem/stromal cells (hASCs) can differentiate into a large broad of specialized cell types to ensure homeostasis of tissues. Thus, they are drawing increasing attention in cell therapy and regenerative medicine. However, culture conditions are still critical to ensure their regenerative capabilities. Here we compared Standard conditions (αMEM containing 10% fetal bovine serum) and Endothelial cell Growth Medium 2 (EGM2) containing few serum (2% v/v) for expansion of hASCs. Both types of hASCs were exhaustively characterized by high throughput studies, Gene Set Enrichment Analyses (GSEA) and differentiation potentials experiments. EGM2-hASCs showed enhanced multipotency and their phenotype was more immature than Standard-hASCs. The adipogenic potential of EGM2-hASCs was clearly more extented, including toward the thermogenic beige adipocyte fate. Data from microarray and GSEA highlighted Transforming Growth Factor β1 (TGFβ1) as upstream factor influencing the becoming of Standard-hASCs which is consistent with higher TGFβ1 content in Standard medium. This was associated with nuclear SMAD3 localization and higher expression of its active form in Standard-hASCs. Moreover, these cells were primed into osteoblast, chondroblast and Vascular Smooth Muscle (VSM) lineages at the expense of their adipogenic potential. Their treatment with TGFβ1 receptor inhibitors resulted in a cytoplasmic re-localization of SMAD3 and a decrease of VSM and osteoblastic lineages markers, increasing in turn their beige adipogenic potential. Therefore, TGFβ1 is a key factor committing hASCs toward osteo-chondroblastic and VSM lineages at the expanse of their beige adipogenic potential. Thanks to low TGFβ1 content, EGM2 medium improves the maintenance of uncommitted hASCs with strong beige adipocyte potential. We used microarrays to unravel differential gene expressions between human ASCs cultured in standard conditions or in EGM2 medium.

Project description:Multipotent mesenchymal stromal cells (MSCs) have been shown to promote tissue repair and inhibit inflammatory/autoimmune responses in preclinical and human clinical trials A major problem for MSC-based therapies is the need to expand MSCs in vitro due to the low frequency of MSCs in tissues and the large numbers of cells needed/patient (1-10 x 10^6 cells/kg). The expansion of MSCs is associated with several problems including loss of homing capacity, onset of cellular senescence, decrease in differentiation capacityand susceptibility to genomic instability and malignant transformation, limiting the utility of MSCs as a cell-based therapy. We have recently reported that murine adipose tissue-derived MSCs (mASCs) and human ASCs (hASCs) express glycoprotein A repetitions predominant (GARP)/leucine-rich repeat-containing 32 (LRRC32). GARP binds LAP/TGF-β1 to the surface of mASCs and hASCs, regulating their secretion and activation of TGF-β1 and promoting their immunomodulatory capacity. Interestingly, silencing of GARP in ASCs significantly decreased their proliferative capacity but the mechanisms remain unknown. As the proliferative capacity of MSCs is fundamental for their success in therapy, we have aimed at understanding how GARP modulates the expansion of MSC.
