RNAseq-based bladder cancer subtyping and IPA analysis with the xenografts from C57 BL/6J

Project description:To subtype the Triple gene knockout tumor we generated and assess the similiarities with widely used MB49 cells, TKO cells were injected into bladder wall orthtopically and bladder lumen by catheter intravesically in C57 BL/6J, while MB49 intravesical injection as a control. TKO intravesical tumors, TKO orthotopic tumors and MB49 intravesical tumors were collected and sent for RNA-seq analysis. In results, both intravesical TKO and orthotopic TKO tumors demonstrates transcriptome profiles that closely resembled human basal like muscle invasive bladder cancer using a luminal/basal/neuronal classifier. Principal Component Analysis (PCA) across samples demonstrated that MB49 cells had transcriptome profiles that differed significantly from TKO tumors. Hence, this study suggest that the TKO tumors closely resemble basal like human bladder cancer and MB49 exhibited no expression of urothelial markers.

In vivo CRISPR screens in mouse model of bladder cancer

Project description:To systematcially explore targets that can regulated tumor's response to immune checkpoint therapy, we performed an in vivo targeted CRISPR knockout screen in a mouse model of bladder cancer. Cas9-expressing MB49 cells were transfected with two sub-libraries to make two cell pools. We first validated full representation of sgRNA composition in cell pools by comparing to that in the plasmid pools. For in vivo screen, we first compared PBS or anti-PD-1 treated tumors to original cell pools to get condition specific change, then compared anti-PD-1 treated tumors to PBS treated tumors to identify genes that may regulate anti-PD-1 response.

Expression data from mice bladder with MB49-induced cancer treated with alpha1-oleate.

Project description:Potent chemotherapeutic agents are required to counteract the aggressive behavior of cancer cells and patients often experience severe side effects, due to tissue toxicity. This study addresses if a better balance between efficacy and toxicity can be attained using the tumoricidal complex alpha1-oleate, formed by a synthetic, alpha-helical peptide comprising the N-terminal 39 amino acids of alpha-lactalbumin and the fatty acid oleic acid. Bladder cancer was established, by intra-vesical instillation of MB49 cells on day 0 and the treatment group received five instillations of alpha1-oleate (1.7-17mM) on days 3-11. A dose-dependent reduction in tumor size, bladder size and bladder weight was recorded in the alpha1-oleate treated group, compared to sham-treated mice. Tumor markers Ki-67, Cyclin D1 and VEGF were inhibited in a dose-dependent manner, as was the expression of cancer-related genes. Remarkably, toxicity for healthy tissue was not detected in alpha1-oleate-treated, tumor bearing mice or in healthy mice or rabbits, challenged with increasing doses of the active complex. The results define a dose-dependent therapeutic effect of alpha1-oleate in a murine bladder cancer model.

Project description:We developed a UPPL (Upk3a-CreERT2;p53f/f;Ptenf/f;Rosa26LSL-Luc) mouse model of bladder cancer and compared it with the existing BBN (N-butyl-N-(4-hydroxybutyl)nitrosamine) mouse model of blader cancer. We cultured UPPL and BBN primary tumor cells as cell lines along with MB49 cancer cell lines and KT immortalized normal urothelial cell lines and implanted them back into mice as cell-line derived tumors.

The expression profiles of these KO cells were compared with non-KO cells also treated with IFNα and infected with ISAV or IPNV.

Project description:A number of studies have reported evidence of positive or negative contributions of galectin-9 (gal-9) to human and experimental malignancies. Some clinical observations and in vitro experiments suggest that cell-associated gal-9 has anti-metastatic effects. On the other hand, extra-cellular gal-9 consistently enhances tumor immune escape. The aim of this experiment is to study the impact of gal-9 gene invalidation in the murine bladder carcinoma cell line MB49, in vitro. It complements the results of : 1) a RNAseq analysis performed on in vitro cell cultures ; 2) a comparative experiment carried out using the same cells as part of a serial transplantations protocol, whose results are already available on Annotare (E-MTAB-9570 ; E-MTAB-9220).

In vivo macrophage engineering reshapes the tumor microenvironment leading to eradication of liver metastases [Visium]

Project description:Liver metastases are associated with poor response to current pharmacological treatments, including immunotherapy. We describe a lentiviral vector (LV) platform to selectively engineer liver macrophages, including Kupffer cells and tumor-associated macrophages (TAMs), to deliver type I interferon (IFNα) to liver metastases. Gene-based IFNα delivery delays the growth of colorectal and pancreatic ductal adenocarcinoma liver metastases in mice. Response to IFNα is associated with TAM immune activation, enhanced MHC-II-restricted antigen presentation and reduced exhaustion of CD8+ T cells. Conversely, increased IL-10 signaling, expansion of Eomes CD4+ T cells, a cell type displaying features of type I regulatory T (Tr1) cells, and CTLA-4 expression are associated with resistance to therapy. Targeting regulatory T cell functions by combinatorial CTLA-4 immune checkpoint blockade and IFNα LV delivery expands tumor-reactive T cells, attaining complete response in most mice. These findings support a promising therapeutic strategy with feasible translation to patients with unmet medical need.
