Campylobacter jejuni interactions with Acanthamoeba castellanii

ABSTRACT: To investigate the interaction of intra-amoebal C. jejuni with the transient host A. castellanii. We then performed gene expression profiling analysis using data obtained from RNA-seq of control and intra-amoebal C. jejuni.

Project description:The persistence of Campylobacter jejuni in the natural environment, despite its microaerophilic nature, continues to be a subject of active investigation. Its survival and persistence likely stem from a combination of factors, including interactions with amoebae. C. jejuni transiently interacts with Acanthamoebae and this is thought to provide protection against unfavourable conditions and prime the bacteria for interactions with warm-blooded hosts. Acanthamoebae play vital roles in ecosystems by preying on microorganisms and establishing ecological niches with various bacterial species, some of which are clinically important pathogens. To gain a better understanding on the role of Acanthamoebae in the persistence of C. jejuni in the natural environment, we examined their interactions at the molecular level.

Project description:Through RNA-sequencing we compared A. castellanii infected with wild type L. pneumophila to A. castellanii infected with an isogenic dotA mutant strain, which is unable to replicate intracellularly. We identified clear morphological and transcriptional changes in A. castellanii over the course of infection. Genes differentially expressed during infection fell into fundamental biological processes, such as small GTPase signalling, cytoskeleton dynamics, metabolism and cell proliferation. Members of the Sirtuin family of class III histone deacetylases were upregulated by A. castellanii throughout wild type L. pneumophila infection. sir6f expression was important for A. castellanii proliferation during infection and L. pneumophila replication

Project description:A culture of Acanthamoeba castellanii cells was infected with mimivirus. At specified times after infection, infected cells aliquots were subjected to total RNA extraction. Cy3 labeled cDNA was hybridized on custom designed Agilent oligonucleotide tiling microarrays covering both strands of the mimivirus genome.
