RNA-seq from mice CD4+CD25+ splenocytes

ABSTRACT: To better understand the function of CD4+CD25+ splenocytes from different ages, target cells from young(8-week old) aged(18-month old) were sorted with magnetic beads. We then performed gene expression profiling analysis using data obtained from RNA-seq of different cells from different age.

Project description:Here we compare the effects of stimulation on cord blood derived CD4+ CD25+ (Treg) and CD4+ CD25- (Thelper) cells, isolated by MACS protocols & expanded in vitro using dynabeads. Expansion was carried out at a ratio of 3 beads/cell in the presence of 1000units/ml of recombinant human IL2 for 8 days, followed by 3 days of culture without beads. RNA was extracted from resting cells on day 4 after expansion. Stimulated cells were treated with Ionomycin for 2hrs prior to RNA extraction.

Mouse splenocyte proteome - Proteome characterization of splenocytes from an Aβpp/ps-1 Alzheimer's disease model

Project description:Splenocytes from 14-month Alzheimer's Disease mouse model were separated with CD90.2 magetic beads into two cell subsets: CD90+ and CD90-. Proteome for these two subset were characterized with offline SCX-nanoLC-MS/MS. MS data was analyzed with Proteome Discoverer 1.3 software. MS/MS spectra were searched against the IPI mouse database (August 16, 2012, 59 534 sequences). SEQUEST search parameters were as follows: two maximum trypsin miscleavages; precursor and fragment mass tolerances of 10 ppm and 0.8 Da, respectively; carbamidomethyl modification on cysteine was a static modification; oxidation of methionine was a dynamic modification. Decoy database searching was employed to control the false discovery rate (FDR) and generate lists of peptides with high (FDR < 0.01) and medium (FDR < 0.05) confidence.

CD38-NAD+ Immuno-Metabolic Axis Regulates Potent Anti-Tumor T Response

Project description:Spleens from the B6 mice were isolated and single cell suspension was made. CD4 T cells were purified from the splenocytes using magnetic bead separation. Briefly, Splenocytes were incubated with biotinylated antibody cocktail consisting of antibodies (Biolegend) to CD19, B220, CD11b, CD11c, NK1.1, Gr1, CD25 CD8. After a wash step, cells were incubated with streptavidin conjugated magnetic particles (BD Biosciences). After washing, CD4 T cells were isolated by applying a magnetic field and removing the untouched cells. Purified CD4 T cells were then activated with plate-bound anti-CD3 plus anti-CD28 in presence of either Th1 or Th17 or Th1/17 polarizing condition for 3 days. Total RNA from the 3 days differentiated Th1, Th17 and Th1/17 cells was isolated using mirVana miRNA isolation Kit (Invitrogen).

Differences in PVAT mRNA profiles of recipient mice transplanted with splenocytes

Project description:Recent investigations raise the possibility that senescent immune cells may fuel vascular damage by perivascular adipose tissue (PVAT) dysfunction, as immune cell infiltration in PVAT increases substantially in the development of vascular disease compared to the healthy state and leads to changes in PVAT-derived effector cytokine levels that comprise vascular biology.Though evidence revealed possible cross-talk among immune cells, PVAT and vasculature, it is less clear if senescent immune cells or a particular cell subset may induce PVAT dysfunction and further impair vascular biology during type 2 diabtes (T2DM). NLRP3 inflammasome plays a critical role in inflammatory response as a major component of innate immunity, it provides a molecular platform that responds to a variety of stimuli, initiating sterile inﬂammation cascade.Furthermore, there is evidence that NLRP3 inflammasome can be activated by sensing danger-associated molecular patterns (DAMPs), which in turn promotes age-related thymic demise and T cell senescence, implicating its potential role in immunosenescence. Here, we employed WT and Nlrp3 KO mice to develop T2DM, and implanted the splenocytes isolated from WT, Nlrp3 KO, WT-T2DM, Nlrp3 KO-T2DM mice that were modeled for 4 months to 3-month-old WT mice. Then, we explored the differentially expressed genes of PVAT in recipient mice implanted with splenocytes and evaluate immune infiltration of PVAT.
