Effect of PDGF, TGF-β and miR-221 on gene expression during proliferation and differentiation of human aortic smooth muscle cells

ABSTRACT: To investigate the key molecular and mechanism during the proliferation and differentiation of human aortic smooth muscle cells, we treated HASMC cell lines with PDGF, TGF-β and miR-221.We then performed gene expression profiling analysis using data obtained from ONT long-read RNA-seq of 4 different treatment.

Transcription profiling of rat aortic smooth muscle cells after PDGF-BB treatment

Project description:Unlike other terminally differentiated cell types, vascular SMCs display remarkable phenotypic plasticity. The adult, differentiated state is traditionally defined by expression of well-characterized SMC contractile genes. Extracellular cues, however, can induce contractile SMCs to remodel toward a synthetic state characterized by a spectrum of proliferative, migratory, and inflammatory phenotypes. We used whole-genome expression arrays to to identify genes associated with SMC phenotypic modulation. Experiment Overall Design: Rat aortic SMCs were serum-starved for 72 hours and subsequently treated with either PDGF-BB or its respective vehicle (n=2). RNA samples were hybridzed to Affymetrix arrays with the intent to identify early genes associated with SMC phenotypic modulation.

RNA Seq Transcriptomic profiling of PDGF-stimulated primary human aortic smooth muscle cells treated with ORAI1 inhibitor relative to controls

Project description:Platelet-derived growth factor (PDGF) signalling and the subsequent activation of the calcium ion channel, ORAI1 are critical drivers of pathological remodelling of native vascular smooth muscle cells to proliferative state, which is a process associated with various vascular diseases. This study aims to reveal transcriptional networks altered following ORAI1 inhibition in vascular smooth muscle cells. To study the effect of ORAI1 inhibition on VSMC biology, we performed RNA-Seq analysis of PDGF-stimulated primary human aortic smooth muscle cells treated with either ORAI1 inhibitor, (n=4) or with vehicle (n=4), and investigated the effect of ORAI1 inhibition on the transcriptional response of cells.

Transcription profiling of human aortic smooth muscle cells exposed to hypoxia

Project description:Systemic arterial smooth muscle cells are exposed to a broad range of oxygen concentrations under physiological conditions. Hypoxia can modulate the proliferative response of smooth muscle cells leading to speculation about its role in vasculogenesis, vascular remodelling and the pathogenesis of arterial disease. The effect of hypoxia has been inconsistent, however, with both enhanced proliferation and growth arrest reported. Nevertheless, these reports support an important effect of hypoxia on smooth muscle cell proliferation and, given its physiological and clinical relevance, this requires clarification. We posited that variation in O2 concentration, within the range that exists in vivo, may have different effects on the proliferation and survival of vascular smooth muscle cells. Experiment Overall Design: Human aortic smooth muscle cells (HASMC) were propagated to passage 6 in SMGM-2 medium reached 80% confluence, the media was changed and the cells were incubated for a further 16 hrs or 48 hrs under either normoxic or hypoxic conditions (1% and 3%O2 ).

SILAC analysis of PDGF-treated primary bladder smooth muscle cells

Project description:SILAC analysis of human primary bladder smooth muscle cells treated with platelet-derived growth factor for 0, 4, and 24 h. Platelet-derived growth factor-BB (PDGF-BB) is a mitogen and motogen that has been implicated in the proliferation, migration and synthetic activities of smooth muscle cells (SMC) that characterize pathologic tissue remodeling in hollow organs. To explore the signals induced by PDGF on a global scale, we performed expression profiling and quantitative proteomics analysis of PDGF-treated human visceral SMC. 1695 genes and 241 proteins were identified as differentially expressed in PDGF-treated primary bladder SMC versus non-treated cells. Analysis of gene expression data revealed MYC, JUN, EGR1, MYB and RUNX1 as the transcription factors most significantly networked with upregulated genes; DDIT3, NFAT5, and SOX5 were most networked with downregulated genes. For protein identification and quantification, raw mass spectrometric data were analyzed with MaxQuant software (version 1.0.13.13). The parameters were set as follows. In the Quant module, SILAC triplets was selected; oxidation (M) and acetyl (Protein N-term) were set as variable modification; carbamidomethyl (C) was set as fixed modification; concatenated IPI human database (version 3.52) (74,190 forward sequences and 74,190 reverse sequences) was used for database searching; all other parameters were default. Tandem mass spectra were searched by Mascot (version 2.2.0.4) (Matrix Science, Boston, MA). In the Identify module, all parameters were default, except that maximal peptide posterior error probability was set as 0.05. False discovery rates for protein and peptide identifications were both set at 0.01.

Transcription profiling of wild type and Hey2 knockout mouse aortic smooth muscle cells treated with PDGF for different durations to identify differentially expressed transcripts.

Project description:The cardiovascular restricted transcription factor CHF1/Hey2 has been previously shown to regulate the smooth muscle response to growth factors. To determine how CHF1/Hey2 affects the smooth muscle response to growth factors, we performed a genomic screen for transcripts that are differentially expressed in wild type and knockout smooth muscle cells after stimulation with platelet derived growth factor. We screened 45101 probes representing more than 39,000 transcripts derived from at least 34,000 genes, at eight different time points. We analyzed the expression data utilizing an algorithm based on Bayesian statistics to derive the best polynomial clustering model to fit the expression data. We found that in a total of 9827 transcripts the normalized ratio of knockout to wild type expression diverged more than 3 fold from baseline in at least one time point, and these transcripts separated into 17 distinct clusters. Further analysis of each cluster revealed distinct alterations in gene expression patterns for immediate early genes, transcription factors, matrix metalloproteinases, signaling molecules and other molecules important in vascular biology. Experiment Overall Design: WT and Hey2 knockout mouse aortic smooth muscle cells were treated in parallel with PDGF and harvested at successive time points for RNA extraction and hybridization on Affymetrix microarrays. We sought to identify differentially expressed transcripts through Bayesian statistical methods that would explain the differential response to PDGF observed in vitro.

The tRNA derived fragments (tRFs) expression profiles in human aortic smooth muscle cells (HASMCs)

Project description:To explore the potential involvement of tRNA derived fragments (tRFs) in the human aortic vascular smooth muscle cells (HASMCs), we conducted tRFs profiling in three pairs of differentiated HASMCs (treated with PBS) and dedifferentiated HASMCs (treated with PDGF-BB 50 ng/ml) by microarray. Our results showed that tRFs were aberrantly expressed in dedifferentiated HASMCs compared with differentiated HASMCs and provided potential targets for novel insights into VSMC differentiation or vascular remodeling diseases.

Project description:Atherosclerosis is the most important mechanism of cardiovascular and cerebrovascular diseases and has become one of the most serious diseases that threaten human health. Vascular smooth muscle cells (VSMCs) are the main cellular components that constitute the structure of the blood vessel wall. The excessive proliferation and migration are pivotal pathological basis of atherosclerosis, in-stent restenosis, pulmonary hypertension and other vascular proliferative diseases. PDGF-BB is a potent proliferative agent for VSMCs, but the specific mechanism of PDGF-BB signaling pathway involved in biological processes such as proliferation and migration of VSMCs is not elusive.
