The acetabular labrum tissue shows unique transcriptome signatures compared to cartilage and responds to combined cyclic compression and surface shearing


ABSTRACT: Purpose: Knowledge about labrum specific features is scarce. However, labrum specific cell and tissue markers such as extracellular matrix gene expression markers may be required during the development of labrum tissue engineering approaches. The study aimed to compare the basal gene expression of acetabular labrum tissue and articular cartilage tissue. Methods: Bovine hip joints were collected from a local slaughterhouse (food industry). From each of the five hip joints, one tissue specimen of labrum tissue was collected from the thickest labrum region and one tissue specimen of articular cartilage tissue was collected from the adjacent cartilage tissue. RNA was extracted by chloroform phenol extraction and purified using the RNeasy Mini Kit from Qiagen. RNA sequencing and functional analysis was performed at the Functional Genomics Center Zurich (FGCZ). Results: More than 6'000 differentially expressed genes were identified. In hierarchical clustering the GO term extracellular matrix organization was enriched and in gene set enrichment analysis the GO term extracellular matrix was enriched. Specifically, COL1A1 and COL3A1 were highly expressed in the labrum tissue and significantly higher expressed than in the articular cartilage tissue. Conclusion: Labrum tissue shows substantial differences in basal gene expression compared to articular cartilage. The here presented expression profile and list of differentially expressed genes may contribute to the development of labrum specific markers.
