Project description:In this study, NAC was UV-crosslinked using the photo-crosslinking amino acid p-benzoyl-L-phenylalanine (pBPA) in order to determine specific interaction sites of the alphaNAC N-terminus in the native protein structure. Furthermore, quantitative chemical crosslinking coupled to mass spectrometry (q-XL-MS) was used to compare crosslink abundances of a NAC mutant versus wild type NAC in order to determine the influence of the mutated motive on the structural dynamics of NAC.

Project description:Reduced reward interest/learning and reward-to-effort valuation are distinct, common symptoms in neuropsychiatric disorders for which chronic stress is a major aetiological factor. Pyramidal glutamate neurons in the basal amygdala (BA) project to various brain regions including nucleus accumbens (NAc). The BA-NAc neural pathway is activated by reward and aversion, with many neurons being monovalent. In adult male mice, chronic social stress (CSS) led to both reduced discriminative reward learning (DRL) associated with decreased BA-NAc Ca2+ activity, and reduced sucrose reward-to-effort valuation (REV) associated, in contrast, with increased BA-NAc Ca2+ activity. Chronic tetanus toxin inhibition of BA-NAc neurons replicated the CSS-DRL effect whilst causing only a mild REV reduction, whilst chronic DREADDs activation of BA-NAc neurons replicated the CSS effect on REV without affecting DRL. This study provides novel evidence that chronic stress disruption of reward processing involves the BA-NAc neural pathway; the bi-directional effects implicate activity changes in BA-NAc reward (learning) and aversion (effort) neurons, with the net overall direction of stress-induced change in activity dependent on on-going stimulus processing and behaviour.

Project description:Endothelial cells, and many other types of cells too, physiologically reside in low O2 environments (~ 2-13% O2 or 15-60 mmHg pO2; [PCN]) relevantly to atmospheric O2 (~ 21% or 160 mmHg pO2 at sea level) in vivo. Such PCN is critical for endothelial functions. The majority of our current knowledge regarding the cellular and signaling mechanisms governing endothelial functions, however, is built on endothelial models established under atmospheric O2 (~21% O2). Herein, we comapred the transcriptional profiles between HUVE and HUAE cells cultured and expanded under PCN (3% oxygen) and standard culture normoxia (21% O2).

NtrBC is a two component regulatory system, nac is a transcriptional activator Mutations in ntrBC and nac result in small plaque formation in Henle cell monolayers compared to wild type bacteria. Gene expression studies were pursued to identify the genes regulated by these transcriptional regulators. A 9 array study using total RNA recovered from the wild type, ntrBC and nac mutant strains in three independent experiments. Bacterial RNA was isolated from infected Henle cells at four hours post-infection. Gene expression from the mutant strains was compared to that of wild type.

Project description:We have used long-term experimental selection over many generations to obtain Drosophila melanogaster strains that can live perpetually in extremely low, normally lethal O2 conditions (4-5% O2). These strains show a dramatic phenotypic divergence from control animals, including a decreased recovery time after anoxia, a higher rate of O2 consumption in hypoxic conditions, and a decrease in body mass due to both decreased cell proliferation and cell size. We used gene expression profiling to identify altered transcript levels in the adapted flies and we show that mutations in many of the corresponding genes confer the ability to survive under extremely low O2 conditions.
