Histone methylation regulates reproductive diapause in Drosophila melanogaster

ABSTRACT: RNA-seq on Drosophila melanogaster ovary (stages 1-7) in: diapause at 12ºC (after 28 days), reproductive at 12ºC (after 28 days), and age-matched control at 25ºC in two inbred genotypes. Each biological replicate is a pool of 25 ovary pairs, and each treatment has three biological replicates.

Next generation sequencing of Coccinella septempunctata L. with non-diapause, diapause, and diapause-terminated states

Project description:The most commonÂ ladybird beetle, Coccinella septempunctata L., is an excellent predator of crop pests such as aphids and white flies, and it shows a wide range of adaptability, a large appetite and a high reproductive ability. In this study, we collected female adults in three different states, i.e., non-diapause, diapause and diapause termination, for transcriptome sequencing. The experimental insects consisted of three different states as follows: Non-diapause female insects were reared at 24Â±1Â°C, with a RH of 70Â±10% and a 16:8 h light: dark (L: D) photoperiod and collected after their first oviposition. Female adults in diapause were reared at 18Â±1Â°C at an RH of 70Â±10% and a 10:14-h (L:D) photoperiod. The experimental diapause insects were collected after 30 days. Diapause-terminated adults were transferred to another climatic cabinet with the 30-day diapause insects and reared under the same conditions as the non-diapause insects. After their first oviposition, the female insects were collected and stored at -80Â°C. Three biological replicates per treatment (non-diapause, diapause, diapause-terminated) were sequenced using Illumina HiSeq 2500.

Gene expression profiling during diapause induction in Culex pipiens mosquitoes

Project description:Investigation of gene expression level differences in Culex pipiens (SB) and C. quinquefasciatus (JHB) at three time points (8, 16, and 24 hours post-exposure) during the early pupal state in standard (25Â°C; 16 h light/8 h dark) and diapause-inducing (18Â°C; 8 h light/16 h dark) conditions. A forty-eight chip study (4 Expr12x135K slides) using cDNA from total RNA collected from two species in the Culex pipiens complex at 3 time-points during the early pupal stage to study gene expression differences between standard (25Â°C; 16 h light/8 h dark) and diapause-inducing (18Â°C; 8 h light/16 h dark) conditions, and between Culex pipiens (diapause) and C. quinquefasciatus (no diapause). Each chip measures the expression level of 18,692 protein coding genes, with 3 probes per gene and two-fold technical redundancy. Each of the 12-plex slides was used for one of four biological replicates. Probes were designed using the C.quinquefasciatus CpipJ1.2 geneset in VectorBase.

Genome wide gene-expression analysis of facultative reproductive diapause in the two-spotted spider mite Tetranychus urticae

Project description:Investigating essential physiological processes in diapausing mites by analyzing genome wide gene expression changes using custom-built microarray. We investigated the molecular biology of facultative reproductive diapause in the chelicerate Tetranychus urticae (Acari: Tetranychidae) by analyzing genome-wide gene expression differences in diapausing and non diapausing T. urticae, using an Agilent custom-built two color gene expression microarray. Analysis of this dataset showed that a remarkable number, 11% of the total number of predicted T. urticae genes, were differentially expressed. Gene Ontology analysis revealed that many metabolic pathways were affected in diapausing females. Genes related to digestion and detoxification, cryo-protection, carotenoid synthesis and the organization of the cytoskeleton were profoundly influenced by the state of diapause. We also further confirmed the importance of horizontally transferred carotenoid synthesis genes in diapause and different color morphs of T. urticae. We made one comparison: diapausing mites (DIA) vs non-diapausing mites (NON-DIA), in 4 replicates. Both types of mites belonged to the T. urticae LS-VL strain. In this strain, approximately 30% of mites enter diapause under the experimental conditions applied. Hence, we were able to sample RNA of mites with similar genetic background that were reared under identical environmental conditions. The labeled cRNA samples were pooled and hybdrized to a custom Sureprint genome wide G3 Gene Expression 8x60K microarray. Data was normalized by Agilent Feature Extraction software (using protocol GE2_107_SEP09). Genespring software (Agilent technologies) was used for the statistical analysis of the data.

Mating precedes selective immune priming which is maintained throughout bumblebee queen diapause

Project description:Diapause is a key life-history event characterised by arrested development, suppressed metabolism and increased stress tolerance. This state allows an organism to avoid prolonged periods of harsh and inhospitable environmental conditions. For species where only mated female adults undergo diapause, mating is performed prior to diapause. Mating can have a profound effect on the behaviour and physiology of females resulting in changes to the expression of key biological processes, including immunity. However, our understanding of how mating impacts long-term immunity and whether these effects persist throughout diapause is currently limited. Here we explored proteomic changes in the haemolymph of the ecologically important pollinator, Bombus terrestris. B. terrestris queens mate prior to diapause (a non-feeding arrest of development that can last 6-9 months). Using mass-spectrometry-based proteomics, we quantified changes in the pre-diapause queen haemolymph after mating, as well as the subsequent protein expression of mated queens during and post-diapause. Our results provide clear molecular evidence for the consequences and benefits of mating at the immune level through the selective increased abundance of antimicrobial peptides that are sustained throughout diapause. In addition our results provide novel insights into the molecular mechanisms by which bumblebees prepare for, survive, and recover from diapause, insights that may have implications for our general understanding of these processes in other insect groups.
