Neferine treatment lower blood pressure in SHR

ABSTRACT: The goals of this study is to compare the differently expressed genes in abdominal aorta tissue of WKY and SHR as well as differently expressed genes in the abdominal aorta tissue of SHR with or without neferine treatment. The rat (n=15) were randomly divided into 3 groups: WKY,SHR, and SHR + neferine - H (high concentration) groups (n=6 for each group). Rat in WKY and SHR groups were intragastrically with double distilled water (dd H2O); while rat in SHR + SHR + neferine - H groups were intragastrically with 10mg/kg/D of neferine for 10 weeks. Then the abdominal aorta were used to identify differentially expressed genes among different groups.

The SHRs (n=12) were randomly divided into 2 groups: SHR, and SHR +ETH (3.5mg/kg/d) groups (n=6 for each group). WKY rats (n=6) were set as control. Rats in WKY and SHR groups were intragastrically with double distilled water (dd H2O), while rats in SHR + ETH (3.5mg/kg/d) group were intragastrically with 3.5 mg/kg/d of ETH for 10 weeks. Then the vascular vessel tissues were used to identify differentially expressed genes among different groups.

Mouse in Control and Ang II groups were intragastrically with double distilled water (dd H2O); while mice in Ang II + neferine (1 mg/kg/day) group were intragastrically with 1 mg/kg/day of neferine for 6 weeks. Then the abdominal aorta tissues were used to identify differentially expressed genes among different groups.

Project description:We examined gene expression profiles in the rat mesenteric arteries using genome-wide microarray technology, and determined gene expression profiles in 3 rat strains and treated ones: spontaneously hypertensive rats (SHR), stroke-prone SHR (SHRSP), and normotensive WKYs treated with 2K1C or 1K1C, or without operation for 3 weeks at 6 weeks of age. To identify candidate genes involved in the genesis of hypertension in the SHR strains in comparison to renal hypertensive WKY, we compared the gene expression levels at 6 weeks of age, isolated genes showing a more than 4-fold increase or a less than 1/4-fold compared with WKY treated with 2K1C, 1K1C or none.
