Differential Renal Gene Expression in Pre-hypertensive and Hypertensive SHR

ABSTRACT: This study examined the renal gene expression profiles between spontaneously-hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats at a pre-hypertensive stage (3 weeks of age) and hypertensive stage (9 weeks of age). In addition, age-related changes in gene expression patterns were examined from 3 to 9 weeks in both WKY and SHR. Five to six individual kidney samples of the same experimental group were pooled together and quadruplicate hybridizations were performed on the NIEHS Rat v2.1, 2.2 and 3.0 arrays.

Gene expression profile of abf1-1 mutant at 36 degrees C

Project description:The abf1-1 mutant and wild type cells were grown at 30 degrees C in YPAD until OD600 reached 1.0-1.2. The same volume of 42 degrees C preheated YPAD was added for each flask. After 45 min incubation at 36 degrees C, cells were harvested and RNA was isolated. Four repeats of microarray results are in this series.

Transcription profiling of ray aortic tissue from different hypertensive models

Project description:To determine if there exists a consistent gene signature associated with vascular hypertrophy among different rat hypertensive models: treated and untreated Wistar Kyoto (WKY) rats and treated and untreated Spontaneous Hypertensive Rat (SHR) rats. Experiment Overall Design: Two strains of rat were studies: WKY which are normotensive and SHR which are hypertensive. Aortic tissue was taken from 6 untreated WKY rats and 6 WKY rats treated with angiotensin. Aortic tissue was taken from 6 untreated SHR rats, 6 treated with nifedipine for two weeks and 5 treated with nifedipine for two weeks and then washed out for three weeks. There was a total of 29 samples. A pool of RNA from 6 untreated WKY rats was used as a reference. Hybridizations were performed in duplicate with a dye-flip and always with the reference as the second color.
