Gene expression profile of lung tissues comparing between ISM1-/- and wildtype mice.

ABSTRACT: Isthmin 1 (ISM1) is a secreted 60 kDa protein that exhibits pro-apoptotic and anti-angiogenic functions in experimental settings. We generated ISM1 knockout mice (ISM1-/- ) in C57BL/6J background through CRISPR gene editing technology to understand the physiological function of endogenous ISM1. The ISM1-/- mice presented spontaneous pulmonary emphysema and immune infiltration under the ambient environment. These findings suggest that ISM1 might be an anti-inflammatory protein important for maintaining lung homeostasis. To investigate the role of ISM1 in the lung under steady state, we compare the gene expression in the ISM1 knockout lungs with that of the wild type. Lungs of young adult mice (2 months old) from wild type and ISM1-/- mice were harvested and bulk RNASeq was performed.
